Obviously, everyone's list will be different.

My list:

1. I-495 (MA)
2. MA 110 (the main route the bus takes for my commute to UMass Lowell)
3. I-93 (MA)
4. MA 213 (short freeway, for those unfamiliar with the area)
5. MA 113
6. I-95 (big gap between #5 and #6)
7. MA 128 (I'm usually, but not always, on the segment where I-95 and MA 128 are together, so they have consecutive spots, but not identical)
8. MA 114
9. MA/NH 28 (MA 28 would remain #9 if MA 28 and NH 28 were considered separate, but NH 28 would fall far behind)
10. MA 125 (is also NH 125, but that segment ranks far down)

I'm not sure whether to construe this thread as "at the present time," "over the course of your lifetime," or at some particular point(s) during your life. I'm also inclined not to count neighborhood streets because obviously the street on which you live at any given time is probably the "most important" in the sense that you use it every time you go somewhere (at least most people would, although I know there are people who have an address on one street but access it via a driveway or alley from another street). In my case, if we go anywhere, via any means (drive, walk, bike, whatever) we start on our street and then turn onto another neighborhood street, although if we walk or bike it's not necessarily the same one because there's a path through the woods (driving, there's only one way in and out of our neighborhood unless you illegally drive on a sidewalk).

So I will interpret the thread as "non-neighborhood routes that have held particular importance since you were old enough to drive."

US 101 for N/S and SR's 42, 38 and 126 for E/W from my home area are the most important.  I-5 is over 80 miles away.

US 101 is THE backbone of the Pacific coast from Bandon OR to Willits CA.  Industry, tourism and local traffic would be gone or severely crippled without it being in functioning condition in BOTH states.

Given this thread started out as a Top 25, I'd figured that I would give it some thought about what roads have had the most impact on my life so far to date.  So in no particular order.....

US Routes

1.  US 66:  Really above all routes this is the one I've probably explored the most in terms of looking for former alignments and basically anything that would be considered "roadgeeking."  I may have taken the Interstates to reach Arizona from the mid-west but all the things I remember were spending time with my father leaving Chicago and traveling the country while seeing the old highway one stub at a time.  Really 66 may not on paper but it really is the road that stands of test of time and refuses to die for a reason...for me it was the gateway to the west and a less mundane existence.
2.  US 27:  From Michigan when I lived on the route to when I used it in lieu of Florida's Turnpike US 27 has remained ever present throughout the years.
3.  US 1:  When I was living on the Florida Keys US 1 was THE road...I really go to know the modern highway and every iteration back to when FL 4a was still around.
4.  US 191:  Kind of odd pick I'm sure to a lot of people but I've pretty much (I think) clinched US 191 over the years.  From the former segments of US 666 through the Coronado Trail north to Yellowstone and Montana there are few US Routes that really take you through such much Americana.
5.  US 101:  Simple...I've driven the entirety of the route and it really is one of the more common highways I've been on.
6.  US 60:  US 60 was a common travel route through my days working all over the southeast.  I know pretty much every mile that existed from California through Texas.
7  US 395:  Another excellent scenic highway traversing the Sierras and Cascades, I've often found myself in the corridor on many a trip to the mountains over the years.

Interstates

8.  I-10:  I've used almost the entirety of I-10 on two previous cross-country moves.
9.  I-75:  This was the route of choice for family trips between Michigan and Florida when I was a kid.  Alligator Alley was frequent foray in my adulthood.
10:  I-95:  As much as I hate this Interstate it did take me from Connecticut to Florida many a time over the years.
11:  I-17:  I can't even count how many times I've driven this route over the years in full between Phoenix and Flagstaff.

State Routes

12.  M-22:  My sister and brother-in-law have a cabin on M-22...if you are from Michigan you'll understand the significance of that.
13.  AZ 101: This was my route of choice for many years when it opened completely in the Phoenix Area.
14.  AZ 51:  I used to live a quarter mile off this route and used it every day for work.
15.  AZ 87:  I used this route along with AZ 260 for frequent work trips up to the Mogollon Rim.  I've must have passed Mount Ord a couple hundred times over the years...
16.  AZ 260:  Ditto AZ 87.
17.  AZ 88:  One of the first routes that I actually did a lot of road research into given the odd nature with the dirt segment, really it played a big part in getting me into this hobby.
18.  AZ 89A:  Ditto AZ 88 along with the fact that it was a former US Route which traversed Sedona and Jerome.
19.  FL 50:  I've traveled this route several times a week from Orlando visiting my Mom in the Tampa area during the years she had cancer.
20.  CA 41:  Yosemite is by far the National Park I've visited the most frequently.  I might not like 41 all that much north of Fresno but it has taken to me to Yosemite countless times.
21.  CA 1:  All the way back to 1993 during a family trip to Big Sur from San Francisco CA 1 has been present.  I don't recall very bad memories along the highway but rather lots of great times...especially spending them with my parents when I was younger.
22.  CA 120:  Go see the Tioga Pass Road thread....the route is beautiful and something I've traveled with a great deal of frequency.
23.  CA 198:  Along with the Generals Highway this is probably one of the more common roadways in California that I clinch on a regular basis given there are two national parks within close access.
24.  CA 99:  As much as I hate 99 it really is just as if important to the Central Valley if not more so than I-5.
25.  CA 180:  Many a foray into Kings Canyon has been taken along the Kings Canyon Highway over the years...not my favorite but among the most frequent for me.

Here I come with exotic stuff.

1. A-23. Almost all the times I'm on the road I hit it.
2. Z-40, Zaragoza's beltway.
3. N-330. Entering Zaragoza from the North, also the road to the Pyrenees (but that is slowly being replaced by A-23).
4. A-2, simply because I bump into it too many times. It has a concurrency with the Northern part of Z-40.
5. A-22, far behind. However several trips a year involve it.
6. N-240, just to reach A-22, which is not completed to A-23 yet. Will be out the list once the one above is completed (due by 2020 they say now).

Everything else is of little importance for me.
Supporter of the construction of several running gags, including I-366 with a speed limit of 85 mph (137 km/h) and the Hypotenuse.

Please note that I may mention "invalid" FM channels, i.e. ending in an even number or down to 87.5. These are valid in Europe.

At present:

CA 87: the absolute closest means of limited-access egress for me from both home and work.
I-280/I-680:  the principal ways "out of town" to the NW (toward S.F.) and north (East Bay).
US 101:  As above, but to the southeast (Morgan Hill, Gilroy area); closest to work as well.  Also alternates northward as alternate to I-280 toward S.F.
CA 85: How to get to the above from home (both directions).
I-880:  Getting to main subcontractor (cabinetry) in Alameda; also record shopping in Berkeley.
I-580:  Getting to business partners/clients in Valley (most often in conjunction with I-205).
CA 17:  Pain in the ass, but only reasonable way to get to friends in Redwood Estates and Scotts Valley.
CA 152:  Main route to SoCal (relatives, friends, etc.).
I-5/CA 99  Once in the Valley, the main routes N & S; which one I use depends upon either destination or time factor -- some things on 99 provoke numerous stops along the way (beerock vendors in Fresno, Bravo Farms in Traver [specialty cheese], etc.).
I-80:  usually use it coming back from Sacramento area, most often late at night when traffic is relatively sparse.

Essentially every other route is used on an as-needed basis; the above are the ones that seem to get used regularly enough to be considered vital.

The first three are practically a three-way tie, since I commute to school on all three of them everyday (my commute is only 20 minutes, but it uses the Conestoga Parkway which has a lot of route number changes for such a short route).

1. Highway 7 - I'm choosing this as my number 1 since it connects Kitchener with Brampton (where I grew up until the start of high school).
2. Highway 85 - I'm putting this above Highway 8 since I drive more mileage of it everyday. I also enjoy how winding it is for a freeway.
3. Highway 8 - The other number in my username! A useful connection to the 401... I also lived on a downloaded portion of it in St. Catharines for a co-op term.
4. The 401 - Almost any big road trip involves taking the 401, as the main route east and west from Kitchener (and really the main highway of Ontario).
5. Highway 6 - The route that takes me to Hamilton and points east. I drove this every weekend during my co-op in St. Catharines, since I would go home to Kitchener to visit my family. It's also the road to Tobermory, one of my favourite summer spots :)
6. The 410 - I haven't driven this in a while, but it was important growing up as the main freeway between Brampton and everywhere else.
7. Highway 10 - Again, I don't drive this often anymore, but it was a main route up to my condo in Thornbury during my childhood.
8. Highway 26 - My condo in Thornbury was quite close to Highway 26 (though that isn't saying much since it's a small town). This was the way to get from the condo to Blue Mountain ski hill.
9. The 407 - I used to use this a lot more in Brampton, but it's still great for trips east where I have to avoid Toronto traffic (though the tolls aren't so nice :ded:)
10. The QEW - The route to visiting my aunt and uncle, and used on many of my road trips to the States. I also thought the view of the Niagara Escarpment was cool, and I wondered what was behind those hills? (apparently nothing much) :-D
11. The Gardiner Expressway - I don't drive into Toronto too often, but the Gardiner is always a highlight (unless it's rush hour :ded:)! The elevated portion between Toronto's tall condo towers is pretty awesome to see!
12. The 406 - I used this route regularly during my co-op term in St. Catharines. The sharp turns in the downtown area are unique for a 400-series highway.

1. I-80 - By far the most important since it makes up most of the route between Davenport and both of my colleges. Also, based on where I live in Davenport, just about every intercity trip (as well as some cross-town trips) will use at least some part of I-80.

2. US 30 - The route I live closest to at the moment (in Ames; also applies to Valpo)
3. I-35

After this, the rankings are arbitrary, but I'll still give it a shot.

I'll skip the routes I've used for commuting (school, work, church, etc.) -- here are some highways farther from home but "close to my heart" I guess:

US 93: I've done the LV - Boulder segment several times, before and after the new bridge, and also around Kalispell, MT and Glacier NP. There's much more of this route to see, but it's nice to have visited more than one part of it.

CA 32: part of a yearly trip to Chico to help out with a fundraiser. Orland has added a few blocks of businesses since the first trip. Chico Farmer's Market is a few blocks off 32, and further east leads you to Lassen Volcanic NP.

US 75: Sioux City to Le Mars, a lot of family in that area. First visits were long before the Sioux City and Le Mars bypass freeways were built.

I-90: driven stretches in WA, ID, MT, SD, MN, IL, IN, OH, NY and MA; visited the "Golden Spike" in Blue Earth, MN where the route was completed (except for Wallace, ID)

US 395: drove a lot of the Eastern Sierra section. Visited the shop (in Bishop) and bought a souvenir cutout sign. Bought the beer (double IPA, not my style. Kept one unopened and shared the rest)

Japan National Route 246: the first signed route I saw on our vacation long ago; then came back to it on a business trip 10 years later and got some fun photos (example)

A303, Britain: not a motorway but does have some expressway sections with interchanges and grade separations. First numbered highway I drove on in the UK. Goes right by Stonehenge. We didn't go in (you can see it from the road); went to Avebury instead.

I-84 (eastern): this is CT roadwork, design, history in a nutshell. Almost all the good parts and bad parts.

FL 293 northbound in Okaloosa Co, FL- the free portion of  the Spence Pkwy tollroad used to bypass the congestion & traffic lights on College Blvd.
I 10 in NW FL- fastest route east & west from home.
