Collectively with Wakulla County Road 61, State Road 61 runs 57.58 miles north from U.S. 98 and the Franklin County line on Ochlockonee Bay northward to the Georgia state line near Iamonia. The southern segment of SR 61 accompanies U.S. 98 north 9.52 miles to the separation with U.S. 319, and U.S. 319 an additional 5.13 miles to become CR 61 east from intersection with CR 368 west at Crawfordville.

CR 61 forms an L-shaped route east and then north to Wakulla Springs, upgrading back to state maintenance at the Wakulla County line. Rejoining U.S. 319 along Crawfordville Road by Lake Munson, SR 61 enters Tallahassee and bisects the city northward to Downtown and the State Capitol. There SR 61 combines with U.S. 27 along Monroe Street before branching northeast along Thomasville Road to rejoin U.S. 319 ahead of Lake Hull. The state road lines Thomasville Road northeast to Bradfordville and Grant County, Georgia as the unsigned counterpart of U.S. 319.

Within Wakulla County, SR 61 and CR 61 travel a rural route with two lanes for the most part. Northward into the city of Tallahassee, SR 61 generally constitutes a commercial boulevard. All of the state road coupled with U.S. 319 north from the capital city carries at least four overall lanes as part of a longer distance route to Thomasville, Georgia.

Construction underway by September 2018 rebuilt the intersection of SR 61 (Wakulla Springs Road) and CR 2204 (Oak Ridge Road) into a roundabout. The state provided $2.7 million for the project earlier that year.1
