U.S. 51 crosses the state of Wisconsin along a 316.6 mile long route from Beloit north through Madison, Portage, Stevens Point, Wausau, Merrill and Hurley. U.S. 51 was superseded by Interstate 39 between Portage and Wausau between 1996 and 1998. The highway north from I-39 to Tomahawk is either a freeway or expressway.

Two business routes for U.S. 51 remain posted in Wisconsin. The route through Rothschild, Schofield and Wausau is fully state maintained. Southward at Stevens Point, the business route there is state maintained from the south city line through Whiting and Plover while the route northward was transferred to the city when the U.S. 10 bypass opened. Two additional loops were dropped from the state system and subsequently decommissioned. A loop through Merrill, following County Q and County K, was removed in 1998. A fourth U.S. 51 Business traveled a rural route through the Lake Nokomis and Tomahawk areas.
