The "most important" stretch of road I haven't been on in MN at this point is probably US 14 between Mankato and New Ulm. I won't be getting that this year, or maybe next year either, as four-laning on that stretch continues.

I haven't come close to clinching the system but the state is so small that I really don't think there is any major route I haven't been on. Maybe MA 213.

I've never been on some Montréal area freeways such as Laval's A-440 and A-640.

The Muskogee Turnpike. It's a diagonal route placed in such a way that it always angles away from wherever I'm going to or coming from. At some point I'm going to have to go out there and do an out-of-the-way backtracking route to clinch it.

The Cherokee Turnpike. The one time I was in the area I bypassed it on what was then US-412 Scenic (now US-412 Alternate).

Of the divided highways in Iowa, I have not been on most of US 61, the Avenue of the Saints south of Iowa City (except for its overlap with US 34), US 30 going east from I-35 to IA 330, and US 20 between US 75 and US 71. I have been on IA 60 when it was a two-lane road but have not been on it as a divided highway from somewhere north of Alton to IA 9.

I have also missed US 14 between Mankato and I-35, mostly because it is easier to get to my cousin's house in Mankato by following MN 109 and MN 22 north from I-90 than to use US 14. US 14 east of Owatonna to Rochester is also a challenge to get to.

Been on all the Interstate mileage in NY and the Hudson River Valley parkways, so has to be one of the parkways on Long Island.

I've been on most of Maryland's. My only missing Interstate is I-370; I also have not been on MD 200.

I believe I've been on - on some portion, not necessarily clinched - of every Interstate in Michigan plus every US route except US-8 (but that can hardly be considered "major").

I've also been on every freeway in the state that has a M number (53, 59, 14, 6).

And I've been on at least most longer-distance state routes, but some only for a short portion of them.

Yeah, I've clinched Indiana's entire highway system, so I was trying to think of what non-highways would count as major roads. There are some like Keystone Pkwy in Carmel and CR 17 in Elkhart County that might be considered major but I've been on those as well. The most major road I can find that I haven't been on any part of is Airport Expressway in Fort Wayne.

There's three Alabama SRs over 40 miles long that I've not driven upon: 63, 106, and 95. Travel Mapping says I have 68 un-travelled routes within the state.

I've completed all the Interstates and been on all the US Routes, save one dinky truck route that's 1.05 miles long.

For Interstates it's easily I-495 on Long Island. For US routes, US 2, US 4 and US 209 are the only ones I haven't been on, so whichever of the latter two you think is more important.

I'm not sure which state routes are significant enough to qualify, but I've been on the handful of cross-state ones that immediately come to mind. At a quick glance, it appears that NY 25 (also on Long Island) is the lowest independently numbered route over 100 miles that I haven't been on at all.

I’ve been on most of my state’s major roads, so I have to really look into it.

For Interstates, I’ve only been on the part of I-195 that overlaps MA 24, so I have been on at least a small part of it in Fall River. The only Interstates in my state I haven’t been on at all are I-391, I-295 and I-395.

I’ve been on all US routes in my state as well, so none of them qualify.

For state routes, I’ll have to go with MA 57. Although I’ve never been on the MA 140 freeway, I’ve been on the surface part of it.

I haven’t fully clinched everything in the Caltrans inventory but I’ve been every state highway (US Routes and Interstates) and probably have completed 95% of the mileage.  I really don’t know what major roadways in cities would qualify as an example for myself.  In a historic context I’ve never been on the Bradshaw Trail in the Sonoran Desert (something I planned to do once with OHV group but it fell through).

For Illinois, just about everything south of Springfield and Champaign I haven't been on, aside from maybe some of I-70 just west of Indiana. I also haven't been on I-180.

For Wisconsin, I haven't really explored anything of the northwest. I've never been on WI-29 if that's something worth mentioning.

Off the top of my head, I'd say I-464, although I'm not sure whether I think that's really a "major road." Otherwise, maybe one of the US highways in far Southwest Virginia (US-19, US-21, US-23, US-421, or US-58 Alternate), but, again, I have trouble thinking of those as "major" in the context of the whole state.

For roads I haven't clinched, it'd be the 417 (I've only been on the portion in Ottawa), Highway 11 and Highway 17 (still have long stretches not done, but hey, they're very long routes)!

For roads I haven't been on at all, those are mainly in Northern Ontario, but the most significant two would probably be Highway 71 and Highway 61 in Northwestern Ontario.

I haven't driven across the bulk of WA 26, which is a major connector between I-90 and the Wazzu campus. I've only been on small sections near I-90 and WA 17.

For Oregon, I have yet to traverse I-84 through the Columbia River Gorge.

I've completed NY*, but the thing with NY is, sometimes there are more major roads that aren't state routes.  This is particularly true in Suffolk County, where it seems like, with the exception of I-495, NY 27, and the parkways, the major county routes are almost uniformly more major than the equivalent state routes.  To that end, I'm going to answer Suffolk CR 111, the connector between I-495 and NY 27 that most/all traffic going to/from the Hamptons and Montauk Point would use if not specifically trying to clinch state routes.

*Technically there are a few asterisks like the couple of blocks NY 9A is signed past its official terminus or US 11 at the border, but they don't change my answer.
