I'm surprised I never listed mine yet. Here's my top 3 in each category:

Interstate:
1. I-81 - From US 211 to I-64 (Staunton).
2. I-95 - From I-495 (Springfield) to VA-30.
3. I-66 - From I-81 to US 29 (Gainesville).
US Highway:
1. US 15 - From VA-299 to VA-230.
2. US 522 - From VA-3 to US 33.
3. US 340 - From Grottoes to Waynesboro.
State Route
1. VA-231 - From SR-621 to US 33.
2. VA-388 - Whole route.
3. VA-253 - Whole route.

To update:

I've since clinched the entirety of MD 206 near Laurel.  Looks like the closet state route is now MD 387 south of Annapolis (Spa Road).  It doesn't really go anywhere interesting so I would probably have make a specific trip there to clinch it.

US Highway:  A piece of US 1 in Baltimore.  In a pretty bad neighborhood; avoided for that reason.  One of these days I'll get it.
State Highway:  MD 387 near Annapolis MD.  Never had a reason to go there.


Another update since I've now been on some of the previously mentioned highways.

Interstate:      I-676 (Philadelphia)  Both PA and NJ segments
US Highway:      A portion of US 1 in Baltimore
State Highway:  A tossup between MD 208 and MD 501 in the Washington DC suburbs

Most of these routes are just in places I would have little reason to go to, save for clinching purposes.  One of these days my OCD will compel me to clinch them.

Update: The closest undriven US route is US-191 where it heads north off US-70 east of Safford AZ, 499 miles away. The closest state highway is NV-267 where it meets Scotty's Castle Rd at the CA/NV line, 419 miles away. When Scotty's Castle Rd is eventually reopened by the NPS, the distance will probably be under 400 miles.

Interesting idea! A few of these stretches were not what I thought they were going to be before looking them up.

Interstate:
(1) I-57, between I-64 and I-70 in IL (approx. 190 mi)
(2) I-49 south of I-44 (approx. 226 mi)
(3) I-35 north of Liberty, MO (approx. 290 mi)
The latter two segments are the only parts of Missouri's interstate highway system I have yet to clinch.

US Highway:
(1) US 61, various stretches in eastern MO that run alongside I-55. Closest single segment I haven't clinched appears to be the bit between Perryville and Jackson (approx. 89 mi)
(2) US 62, between Corning and Piggott, AR (approx. 112 mi)
(3) US 54, between Kingdom City and Mexico, MO (approx. 143 mi)

State Highway: (this one was tricky. I only counted numbered routes here, would be much tougher if I included random dead-end stretches of Missouri's lettered routes even though they are technically "state highways." Still can't totally guarantee I didn't miss one.)
(1) MO 51, either direction from its junction with MO 72 in Bollinger County (approx. 71 mi)
(2) MO 172, east of its junction with US 67 and MO 49 north of Poplar Bluff (approx. 85 mi)
(3) MO 28, north of Dixon (approx. 86 mi)
