Turn the clock back to before the Labor Day Hurricane of 1935 and you would ride the Overseas Railroad extension of the Florida East Coast Railway. Otherwise it'd be hard to fathom getting away from the Florida Keys back to the mainland without the Overseas Highway, boats, and planes.

The actual solution for Boston to Worcester (as intended by Roadgeekteen) is enter I-93 from Science Park MTBA station in order to not touch US 1, then I-495 and I-290.

As for NYC to Poughkeepsie, use Hutchinson River Parkway to I-684 to NY 22 to NY 55.

Now for a tricky one: Rock Springs to Evanston (both in Wyoming) without touching any Interstate.

That's taking the long way sir, and IMO you can touch banned routes without using them (unless a concurrency is required), even if using mass transit. That's why I recommended using Amtrak's Lake Shore Limited (which partially runs on track owned and operated by the MBTA) between Boston's South Station and Worcester's Union Station, or using the MBTA's Framingham/Worcester Line between Boston's South Station and Worcester's Union Station.
