I'm almost certain I've gotten off of every exit on I-35E in Minnesota. I am 100% certain I have all the exits of I-35 clinched south of 35E/W and north of the Iowa border. I've clinched the exits of Minnesota's I-394 as well, and ditto I-190 in South Dakota.

I-80 and I-280 in NJ and NJ 3. I have almost all of the NJ Turnpike exits clinched too. Exit 5 is the only one that I have never used.

The Oak Street Connector (CT 34) and the Mount Carmel Connector (CT 40) up by New Haven. Also, the Loop Parkway on Long Island, if that's not cheating since it has no exits other than the termini.
The only one of serious length where I have gotten off at every exit is I-695 in Maryland, which is 51.46 miles long (if you count both the interstate and state sections) with 43 exits.
I have gotten off at almost every exit of I-95 in Maryland (44/50), missing 6 of them in Prince George's County. I have gotten off at every exit of I-83 from Baltimore to York (33 exits) but am missing a good chunk of exits between York and Harrisburg.
The only one I can think of would most likely be the US 460 Christiansburg-Blacksburg Bypass including the intersections at VA 314(Southgate Drive) and the US 460 Business intersection at the western(or more northern) end of the bypass.

I-74 between its end in Davenport and the Moline Airport. After all, that's as far as the freeway goes, and then 74 splits off.
The US-61 freeway between Davenport and De Witt (thank you, Driver's Ed). (a debatable one is the 61 "freeway" bypass of Blue Grass, IA that has two exits and one overpass).
Easily: I-180 in Pennsylvania. I could go you one better on that one and say with confidence that used every single ramp in all directions multiple times. The same "every ramp" claim goes for I-283 as well, but that's not much of an achievement.
It occurs to me that I've also used every interchange on I-476 in PA–both the Northeast Extension and the Blue Route.
I could almost count the mainline of the Pennsylvania Turnpike (as well as I-76), but I think I'm missing Beaver Valley (PA 8) and the two relatively new E-ZPass only interchanges.
For me it's the Indiana Toll Road, and I-865. :-D (It's only termini are the ends.)
I almost added the St. Joseph Valley Parkway, but I think I'm missing Tabor Rd. in Michigan.

many, but I don't know of any for sure. off the top of my head, I've got a "probably" for I-5, 15, 25, 90, 94... all of 80 west of approximately Joliet. I may be missing a few I-5 in Portland or Seattle, or I-15 in Vegas; generally it's the large cities that throw me, as when I am exploring rurally, I know to get off at every exit to look for old signs, while in the cities it makes more sense to look for the trailblazers stashed 2-3 blocks away on city streets.

I've got all of I-480 in Omaha/Council Bluffs in both directions and all of US 75 in Omaha and suburbs. I believe I have I-129 as well. I also have I-180 in Lincoln.
I-395 in VA/DC.
Also 270Y, 395, and 495X in MD and the Whitehurst Freeway in DC. :-P

If you're heading south, you could just get off the turnpike at exit 5, take 295 south
