First time traveling through those states. Anything to look out for? Quickest route is what I'm looking for.
I'm going to have to go against what all the tourist guides say and not recommend hearing jazz at Preservation Hall in New Orleans. You pay to hear a 3-4 song set. It's really disappointing.

I would imagine straight down I-49/US-71 would be the best route.

Eh, I also feel I also got my 60-minutes worth for $5. Had to wait outside for 30 minutes ahead of time, but hey, you can drink outside and chit chat.
No smoking inside, either.
I-55 works if you'd like to drive on the world's longest bridge? Otherwise, I-49 seems your best bet, although I-10 through Baton Rouge is usually quite congested; west of BR is also full of bridges, except they're more narrow.
Here's how I'd go.
1) Take I-49 / US 71 south to MO 13.
2) Take MO 13 to Springfiled.
3) Get on US 65 and take it south all the way to I-20.
4) Take I-20 east to MS 27. Take MS 27 southeast to I-55.
5) Take I-55 south to New Orleans.

$5? I had to pay something like $15 ($30, since I had a date with me). And it definitely wasn't anywhere close to 60 minutes. More like 30.
ETA: Looks like you can reserve seats now and pay $35-45 per ticket.

Hmmm...for me, it was the summer of 2004: pre-Katrina and off-season.
The reverse of the routing I took would work well. From KC:
1) I-49 South to 7 Highway (Southern end of duplex)
2) 7 Highway South to 13 Highway
3) 13 Highway South to I-44
4) I-44 East to US 65
5) US 65 South to US 60
6) US 60 East to US 63
7) US 63 South to I-55
8) I-55 South to I-10

This is the route I would use as well. You'd have to deal with a curvy, two-lane section of US 63 in northern Arkansas, and I-55 in Mississippi is boring as all hell, but overall it's not a bad trip (I've driven all of this route multiple times, albeit not at once).
