When passing through the New York State Thruway i came accross the Lock 13 Living History Park and Rest Area. And i started to think, is there anything else like this in the country. Where there is a major museum/park that is only connected to a road like the thruway.

Before they closed it, of course.
Not really a Museum but an attraction. Multanomah Falls is off of I-84.

That's not a Space Shuttle, that's a Saturn 1B rocket. :P
The US Space & Rocket Center is technically only accessible by interstate, as you have to go through the interchange at Exit 15 on I-565 to get to it, no matter which road you're coming from, but I'm not sure if it counts.

There's another one in Port Byron for the historic Erie Canal lock. Not sure if it's reopened yet; it was closed so they can construct the building and finish up a few things.
Do the Cabazon Dinos count? Not technically directly accessed from I-10 but I-10 is for all practical purposes the only way to get there.
I'll nominate the Atchafalaya Welcome Center. There are ways to get to it that don't involve I-10, if you have a boat or a big truck.
It's not a museum, but the rest area on I-77 southbound just south of Buffalo, OH has a historical marker about the wreck of the US Navy airship Shenandoah, which crashed during a thunderstorm in September of 1925.

The restored/revamped Midway Service plaza on the PA Turnpike (Eastbound) was done with some museum-esque features.
