For me its US 64. I have seen the east end on the Outer Banks of NC several times, and I passed the west end coming down US 160 from Colorado last December.
Probably I-70 for me. It's certainly the longest road I've clinched.
Longest route I've seen both ends of as a driver is I-84. Longest route I've seen both ends of as a passenger is I-89.
I was going to say I-75. I haven't clinched it (only state in which I've travelled all of it is Florida), but I've been to both ends–the Florida end multiple times, the Michigan/Ontario end on a family vacation in 1987 that saw us visit Isle Royale and then Manitoulin, followed by the ferry to the Bruce Peninsula and on down through Ontario on the way home.
But then fillup420's post prompted me to look up the exact locations of the ends of US-64 and I realized I've been to both ends of that as well, so US-64 it is. Again, not nearly a clinch.
The longest route off the top of my head that I've driven to both ends of is MN-200, all 201 miles of it. I cannot think of any longer state routes (I haven't hit both ends of either MN-1 or MN-23), and there hasn't been any US or Interstate (2di ones, I mean) routes that I've driven to both ends of.
Edit: actually, scratch MN-200. MN-65 wins at this, being 272 miles long.
I-10 which also happens to be my longest route clinch as well.
I-630 in Little Rock.... :-/
I-88 (NY), which I have clinched.
I-81, which I have a complete southbound clinch of and nearly a complete northbound clinch (I'm missing about 80 miles in VA and between I-80 and I-84/380 in PA). Thanks in part to the two three fore-mentioned Interstates, I never have a reason to use that section when traveling to/from home.
Edited to add: I forgot, I have seen both ends of a longer route. I've seen both ends of I-10. I don't nearly have the route clinched, but I have driven the eastern terminus and I've seen the western terminus as a passenger. I've never driven in California. The last time I was in CA I wasn't old enough to drive.
I-80. Clinched the west end (east from terminus to Reno) countless years ago; clinched the east end in 1988 (from I-287 east to its terminus). Did the rest in one fell swoop in the spring of 1989, west-to-east, Sacramento to NJ. Actually, one of the best road trips I ever did; set no speed records -- did it in 4 1/2 days.
US 50. I've driven less than ten miles on the highway in both Sacramento and Ocean City.
I-80 because of several vacations to the NYC area (though I haven't been on it between Cheyenne and New Jersey).
I thought the answer was I-90, but I just learned that it has since been extended in Seattle beyond the western terminus that I drove in 1999.
I've seen the both ends of I-10 and I-40. I've also seen both ends of Route 66 if that counts nowadays ;-)
I've been to both ends of US 64 as well, but certainly don't have it clinched.
I-70, which also happens to be my longest clinch as well.
Notable ones where I do not have a clinch of the entire route:
I-40
US 11
US 30
US 40
US 52
US 64
US 70
US 80
US 89
US 90
Does it have to be signed? Because if not, I think it might be FL 93.
For fully-signed routes, I can't think of anything longer than I-16. Sadly, I took I-295 around Jacksonville last summer instead of going straight through, otherwise I'd have the obvious answer of I-10.
I-90 in MA & WA (nowhere near clinched).
Longest route clinched: I-495 in MA.
I have seen both ends of US-127, but have only traveled the portion from I-75 in Michigan to I-70 in Ohio, and from its junction with US-27 in Chattanooga to the town of Signal Mountain.
I-10 I have seen its east end and west ends even though I have not yet clinched it all. Seen both terminuses of I-75 but need to clinch the interstate between I-94 and US 127 in MI as the rest of it is done.
Also I have seen both ends of I-55, but not yet clinched it, and I-65 I have also seen both ends where I did clinch it through KY and TN and parts of IN and most of AL.
Oh, yes I-85 as I have driven through Petersburg, VA plenty of times on I-95 and I drove I-65 through Montgomery, AL.
I-90 on separate vacations to Seattle and Boston.

I-90 on separate vacations to Seattle and Boston.
Same here, but nowhere near clinching it.
I think US 27 might be the longest north-south route I've seen both ends of. I know I've driven through the "north" end of US 101 in Olympia, but can't quite confirm if I've driven along its southern terminus.
My longest entire-route clinch is I-59, although I've driven much more continuous distance of I-95, I-75; and maybe I-40 and I-20.
Cool topic!
For me, it's probably I-44 (634 miles), and second place might be US-400 (481 miles).
I've really never kept track of this stuff, though, so I might be wrong.
I-76 (East)
I-476
I-85
I-87
US 13
US130
Interstate 84 (east), and in fact going from Mass to PA all at once.
The downfall of driverless cars: A great idea. But what went so wrong?
I-5
I-94
US 101
A trip to Everett would add US 2 (western) to the list
In Canada: ON 7 (535.7 km)
In the states: I-476 (212.6 km) - also clinched :)
I-90 (but not all the middle parts...)
I'd probably have to say (present) I-57. Seen both ends in Sikeston and Chicago's South Side
I'm not sure if I have it clinched, as I can't think of a time I drove from the I-57/I-94/Dan Ryan/Bishop Ford Junction and the I-57/Tri-State/I-294 Junction
Next time I have a car up in Chicago, I will need to find an excuse to drive that short section of I-57. I've ridden the (mostly) parallel Canadian National/Illinois Central tracks on the Amtrak Routes between Carbondale and Union Station, Chicago (Illini, Saluki, and City of New Orleans trains) plenty of times
Close on I-80, but took the Lincoln Tunnel across the River in NY/NJ, not the Washington Bridge. Walked around the Western end of I-80 in Downtown San Fran a few years back
I-55. Still need a few spots in the middle for a full clinch, though.

Same, except I am missing between WA 519 and I-5 (does that count?) Otherwise my interstates are 82 (which is also my only 2di clinch) and western 86.
For US Highways, my longest for sure is US 189 at 322 miles. Honorable mention is US 89, which I am missing the last 10 miles between Babb and Canada (but I don't have the rest clinched).
Like a few others here I have seen both ends of I-90, but that's actually not the longest for me. I have also been to both ends of US 30, which is a few dozen miles longer. And the definitive answer for me since there are only two longer routes in the US (US 6 and US 20), and I have not been to the western end of either of those.
Clinched route: I-35
Non-clinched route: I-25
I was initially thinking it'd be I-476 or the PA Turnpike mainline, if that counts as a "route", but the answer hands down is US 13, though I don't think I've actually been on it at its southern end. I've only passed through on 95 or exited onto 295.
Me?
Likely I-24. Clinched it, too.
In-state, I've also seen both ends of WI 13, WI 16, WI 23, WI 29, WI 32, WI 33, WI 35 and several other of Wisconsin's longer state highways.
Mike
I-80, which I have clinched, and I-40, also clinched.

Seen both ends of I-74 too, not counting that craziness in NC

The longest I've clinched is I-90, but I have seen both ends of US 20, so that's my winner for this category.
As a passenger, would be either I-87, I-91, or US 5.
As a driver, would either be I-295 (including future extension to the PA Turnpike interchange), US 222, or NJ/NY 94. Didn't drive the portion of NY 94 from NY 32 to US 9W, but I saw it on a trip down US 9W.
Seen both ends but not clinched: I-80, though several years apart on a different trips.
Seen both ends and clinched: I-57. I'm close to getting western I-74, only need to get the mainline ramp movements at the two I-465 interchanges.
Oh wait, I remembered I have also seen both ends of US 491. This was on the same trip that I saw the west end of US 64. We took a detour on US 160 to see Four Corners, then made our way back to US 491 to get to I-40.
Ontario Highway 17 (1964 km)
I've clinched about half of it.

I have actually seen every end of I-74 in NC. There are currently 6 "termini" in the state.
Longest route where both ends have been visited: I-90
Longest route clinched: I-64, I believe.
I-84 eastern (232.39 mi) - clinched (entire length)
CA 99 (424.85 mi) - both ends, but not entire length
US 101 (1,540 mi) - both ends
oh wait
US 6 (3,198.87) - both ends
For me the longest route I've seen both ends of is I-65 (887 miles) in Gary and Mobile. I've also clinched it in slow motion, over a period of 40 or so years.
Longer near misses include US 31 (1280 miles) and I-90 (3020 miles). I've seen the north terminus of 31 near Mackinaw City, but didn't go east from Mobile to Spanish Fort. I've been to the west terminus of 90 in Seattle, but haven't made it to downtown Boston. I've not come close to clinching either of those routes.
Seen both ends of I-84 (E), I-395 (CT, MA), NJ 700 (NJ Turnpike), US 113, Taconic State Parkway, Hutchinson River Parkway/CT 15 (including all parkways and the Wilbur Cross Highway), CT/MA/VT 8.

I have been on all of both US 6 and US 20, so those would be on my list. But I suspect (too tired to check out) that the mainline of the Trans-Canada Highway, from Victoria BC to St. John's NL, would be longest of them all for me.
The Pan-American Highway (even just the part north of the Darien Gap) would be a possibility for someone else, but not me since I have very little mileage outside the U.S. and Canada.
I-90 longest seen both ends
runners up:
I-80
I-5
US 101
I-84 western
Depending on how we traveled through Jacksonville during a family vacation in the early 1970s, I may have seen the southern terminus of US 23. I definitely have seen the northern terminus.
I'm thinking the longest north-south route I've definitely seen both ends of is US 25, which I have clinched. (Having been on the portion from Georgia to well into North Carolina on the same aforementioned vacation.)
This is an easy question for me to answer. I-90 in Boston and Seattle.
US Highway: US 12 (2483 mi)
Interstate Highway: I-10 (2640 mi)
In both cases I have actually driven on the road at one end and just crossed the road at the other end.
Indiana state highway: IN 9 (190 mi)
I-55, I-57, I-65, I-75
I've been to both ends of I-10, but hardly anything in between
I-10 (maybe) I have been to the Jacksonville end, and I have been to LA, but not down by the beach where the actual end is.
US 20, US 6, I-94, I-90, I-80, I-70, I-40, I-95...
I-85, I-91, I-89, I-87 (NY), I-81, I-93, I-59, I-65...
...I-30, I-79, I-84 (both)...
I-97 :D
I suppose my list is sad. Longest Interstate I have seen both ends is I-88 in Illinois. The longest US Route is US 218, but I haven't been on any of it, except for the overlap with US 34, south of Iowa City. For state route, I think it is IA 3.
US 101 (1,540 mi), and I can't even think of a remotely close second. I've been to New York City but not quite near I-80's eastern terminus. I've clinched I-84 from Portland to Ogden, UT, but not from there to Echo. I have been to San Diego but not near the Mexican border (though I have been to Canada via I-5, long ago).

I mean, if we're going to assign a beginning and ending to that which doesn't have a physical beginning/ending, then using MM 0 seems to be the most logical answer. So, to answer your question, yes.
You could, however, differentiate it by going past MM 0 in both directions, with traveling clockwise being the beginning and counterclockwise the ending.

I didn't realize until now that all Interstate beltways have a point where mileage resets to zero. I kept trying to find a counterexample (say, where the southern portion is a duplex and uses the other highway's mileage), but I can't find a single one.

Not an Interstate, but Spanish Z-40 around Zaragoza. The Northern part of it overlaps A-2 and uses its mileage. Once it splits it counts from the start of the overlap, so it doesn't really have a kmpost 0.

I've been to the northern end of I-75 several times. And at least one of the former southern ends, but I haven't been to the Miami area since it has been extended to there.
I know that my clinch of I-75 is in doubt since my only E/W crossing of FL on what is now confirmed to be I-75 was on the old Alligator Alley configuration in 1981. That said, there is no doubt that I have been to both ends of I-75, as well as US-31 and US-41.
Since my I-75 clinch is off the table I would have to say I-24 (hard for me to be sure, it seems my stats have been deleted from the Highway Browser page)!

Interstate (as an adult/driving myself): I-275 Cincinnati (never clinched a 2di)--84 miles
Interstate (my whole life, including childhood): I-71--347 miles
US Highways: Only ever clinched Business or Truck Routes--Longest is US 17 Business Myrtle Beach--23 miles
State Highways: OH 126 (Greater Cincinnati)--40 miles
Isn't seeing both ends of I-275 essentially being at any single point on I-275?

I think we've already determined that seeing both ends of I-275 is essentially being at MM 0.

Yeah, you know that little voice in your head that says
You probably shouldn't do that? It's there for a reason.
Now that I thought about this some, it's possible I have seen both ends of I-25. I definitely saw the south end in 2009 when a buddy and I went to Phoenix from El Paso. As for the north end, I was only a young kid then, but we were definitely following the general path of I-90. But I-90 may not have been built yet in the Buffalo, WY area and it's also possible Dad may have simply decided to drive thru town.
As passenger, I-40. As driver, I-81.

Me?
Likely I-24. Clinched it, too.
In-state, I've also seen both ends of WI 13, WI 16, WI 23, WI 29, WI 32, WI 33, WI 35 and several other of Wisconsin's longer state highways.
Mike
Recalling my roadtrips, it is not I-24. Even though I have not 'clinched' it, I have also seen both ends of I-81.
Mike.
