In NJ, CR 533 cannot be followed SB without making a detour, due to it being routed over a one-way road west of Princeton. NJ 29 was also formerly like that in Lambertville, until the route was rerouted in the 2000s, consuming all of NJ 165 in the process.
Are there any other examples of numbered routes that are routed in both directions on one-way roads, or are otherwise unfollowable (due to turn restrictions, etc.)?
MD-7 in Elkton, Cecil County, can only be driven in one direction (I do not remember which way).

No. It is a multi-section route that ends before the one-way segment begins; the one-way segment is not maintained by MDDOT.
VA 399 (unsigned) is one-way for its entire length
VA 131 has a section in downtown Appomattox that can only be driven one way. There is no posted way to get around this segment heading the other direction
The eastbound US 33 to VA 33 transition cannot be driven due to the physical inability to turn left at the intersection this would occur. There is a minimally posted route to get around it.
VA 393 (no longer signed) is one way for some of its routing
I should have thought of this much earlier, but a small stretch of VA-244 in Annandale falls into this category. From its intersection of Backlick Rd and Maple Pl, it is one-way eastbound westbound to its western terminus at VA-236. Traffic headed for VA-244 westbound eastbound is directed to utilize John Marr Dr back to VA-244 west east.

HI 5600 (Kapa'a Bypass on Kauai, signed only with small route number plates under milemarkers) is one-way southbound only for its northernmost ~0.7 mile.
Hennepin CSAH 33 (part of Park Ave.) in Minneapolis. The entire length from 46th St (CSAH 46) to Washington Ave (CSAH 152) is one-way northbound.
Similarly, Portland Ave between those same streets southbound (CSAH 35), but CSAH 35 continues as 2-way traffic south of 46th Ave, whereas CSAH 33 does not.

The south end of PA 72 in Lancaster cannot be accessed SB as the direction ends at US 222/PA 272 (McGovern Ave at Prince St) (https://www.google.com/maps/@40.0533561,-76.3104783,3a,75y,199.02h,98.02t/data=!3m6!1e1!3m4!1s9t-c6a3FpDccX37lUhHKAg!2e0!7i13312!8i6656). PA 72 is NB only (https://www.google.com/maps/@40.0361419,-76.3053272,3a,75y,19.85h,81.85t/data=!3m6!1e1!3m4!1sDUPSViNwLUtV74JxnX9fRA!2e0!7i13312!8i6656) on Queen St as it begins where US 222/PA 272 NB goes onto Church St.
