I recently made another thread about the largest counties without a US route or interstate, but I feel like that thread is a bit inadequate to encompass all the largest counties in general, not just the largest in each state.
I want this thread to be about the US counties above or around 50k residents without a US route or interstate.

Prior to 2015, Indiana had one county greater than 50K that wasn't connected to any interstate or US Highway, and that was Monroe County (pop. 137,974). Now that I-69 is in place and signed through Monroe County, there are no countries in Indiana greater than 50K that aren't connected with an interstate and/or US Highway.

Correct me if I'm wrong here, but I think Wayne County, pop. 90,856, and Fulton County, pop. 53,743, are the only other NY counties to qualify.
Quick rundown of eliminated counties, from smallest population to largest:
Franklin has US 11, Genesee has I-90, Otsego has US 20 and I-88, Columbia has I-90, Washington has US 4, Herkimer has I-90, Livingston has US 20 and I-390, Warren has I-87, Madison has I-90, Sullivan has US 209, Cattaraugus has US 219 and I-86, Cayuga has I-90, Clinton has I-87, Chemung formerly counted but now has I-86, Steuben has I-86 and I-390, and Putnam has US 6, US 9, and I-84.
Everything else is either more populous than Tompkins (which we already know is the most populous NY contender thanks to the other thread), or less than 50K population.
Every county in the state of SC has at least a US route passing through it.
It appears there are no counties that meet the criteria in any of the eight Interior West states (NV, ID, UT, AZ, MT, WY, CO, NM). Partly because there aren’t a whole lot of counties over 50k in those states, and partly because western counties typically cover enough area to include at least one interstate or US highway.
