This is related to the clinching thread. For me, probably I-95 between exits 35 and 36.
I-35 between Exits 106 and 108A. Only river crossing for miles. Used to live south of the river, so I had to use it to go anywhere to the north. Now I live north of it and have to use it to go anywhere to the south or west. It was part of my commute for 11 years.
I-80 between Exit 304 and 307 in Stroudsburg.
Probably the section of I-85 in GA between exit 104 (old exit 40, Pleasant Hill Rd) and exit 106 (old exit 41, SR 316). But since the late 00s, motorists do not enter mainline I-85 when going between those two exits, they use the associated C-D lanes.
Has to be I-75. Now the exact section? I first worked in Highland Park, so commuted south. Then moved to the new HQ in Auburn Hills, so commuted north. I don't recall which location I worked at longest.
Almost certainly I-495 in Virginia, and I presume I could narrow that down to the segment between Route 236 (now Exit 52) and Van Dorn Street (eight miles away, but it's Exit 173) based on where I live now and where I grew up.
I-17 north of AZ 101 to AZ 260.
It's probably close to a tie for me between:
I-435 (KS) between US69 and Metcalf
I-35 (KS) between Lamar and Southwest Boulevard
I-25 (CO) between Evans and University

I-95 between Exit 133 and 148 in Virginia which has been part of both my commutes since 1996
I've moved around so I have have a few. I lived in Michigan the longest
In Michigan, I guess I-75 between exits 34 and 41. Have only been on that section a few times in the past 15 years. I-94 between M-39 and US 24 is close.
In Colorado, I-70 between exits 276 and 282 (CO 2 and I-225).
In California, I-80 between CA 65 and CA 193.
Currently, its I-475 between I-75 and US 24.
I-71 OH between exits 17 and 19. South of there, I continue on I-71 most of the time, but sometimes get off to I-275 in either direction as well.
This is actually harder than I thought.
My first gut reaction was I-91 from MA-9 to I-90 and then to the Berkshire Spur. Then, I-87 to Selkirk (essentially Northampton. MA to Selkirk, NY). Commuted weekly for 8 or 9 months doing that.
But, that is only 36 weeks at the most, or 72 one-way trips.
Living in the Capital District, I frequently used I-90 from Wolf Road to US 9 to get to church. That was probably way more than 72 trips. Not sure how many times I drove on I-787...
And then I lived in MA for over a decade and I'm not sure how often I was on I-91, either getting on at MA 116 in Whately/Deeefield or at MA 9.
If I had to lay a bet, it is probably some length of Free 90 in the Capital District.
When I lived in NC, it was I-795 between Fremont and Goldsboro. I've driven all of I-795 numerous times (even during the short time it was signed as US-117), but as far as regular commutes go, that was the part I drove the most, since I've lived in both places.
Next would be I-95 between Wilson and Rocky Mount.
I-75 between Saginaw and Flint.
Overall I have driven on I-94 more than any other interstate highway. Especially the section east of WI-26 toward the Milwaukee area. But I have also regularly taken it down to the Chicago area and up northwest of Madison.
I-39 between Stevens Point and Wausau.
Though the more time that passes, eventually the 39/90/94 combo from the Beltline to WI 19 may eventually pass it.
(Yeah, yeah 94 isn't on the entire segment but brevity, man.)
I-94 between MN 101 and 494/694.
I-96 between MM 175 and 191, several hundred times in each direction. From 1984 to 1991, this was part of my daily commute from the western suburbs to downtown Detroit.
In State: I-69 MM 305-309 (Illinois Rd/IN-14 to US-30/33/IN-930 in Fort Wayne, IN)
Out of State: I-75 MM 61-110 (I-70 Vandalia to US-33 Wapakoneta, OH)
I-65 in Indiana between exits 6 and 103
Harder to figure out than it appears at first glance.
My best guess would be hidden I-595 in Maryland, between I-95/495 and MD 197, due to my current commute, and often going out to lunch on workdays.
It may be hidden, but it's still an Interstate!
One of the three Interstate approaches to the I-490/I-590/NY 590 junction. Probably I-490 west of the interchange, but almost impossible to say for sure.

I-80 the San Francisco to Sacramento section is the one I am on the most.
I-280, I-680, I-305 and I-780 are the other most used interstates I have been on.
I-5 in Seattle.
I haven't lived there full-time since I was in high school, but it's still probably I-80 along the north side of Davenport, IA. Specifically, the 5-mile stretch between I-74 and the exit I take to get to my parents' place.
I lived in South St Louis City or County for almost 30 years, mostly working near the brewery. I-55 was my main drive to work, the brewery, the good restaurants in Soulard and Midtown, my favorite bars, and the Arch/Downtown. It was also the way to Illinois via the PSB until I moved to SoCo. I drove I-55 between Cherokee and Bates at least 400X/year for about 20 of them and plenty of times other years, so probably close to 10K total drives. I doubt anything else comes close.
I-39/90/94 between US-151/East Washington and US-51/Stoughton Road in Madison.
I-485, the Gov. James G. Martin section.
I-84 between Exit 32 (CT 10; future 46) and Exit 35 (CT 72 East; future 50). From here, I'm usually either going to somewhere near New Britain/Berlin/Newington, or continuing on to the Hartford area.
I-540 Arkansas
I-93 between I-95/128 in Woburn MA and Route 102 in Derry, NH. Had family that lived in Derry for over thirty years, and my dentist and stylist (both old family friends) still have their businesses there.
This is a challenge to figure out because I have lived in 4 different states. I suspect it was probably the section of I-80 between 42nd Street and I-480 in Omaha because when I had my courier job way back in 1996 through 1998, that was the route to downtown Omaha and the airport from my job headquarters, although it wouldn't be unreasonable for it to be I-480 for the same reason. Of course, quitting that job wouldn't make me stop taking those roads. Even moving away wouldn't.
I-35 in Kansas between US 69/Overland Parkway and US 56/69/169/Shawnee Mission Parkway.
I-294 from 95th Street to I-88
I-95 between Richmond and Carmel Church. After that, I use VA 207 over to US 301 into Southern Maryland. Traffic on 95 gets horrible at exit 126.
205, between exits 12 and 19
I 71 from louisville (I 264 ramp to 71 north) to columbus (exit 104 frank road). that was the exit to get to grandma's house. in the years since she died, I've taken I 71 north (that section at least) at least 8 times.
second most is I 65 both ways from the gene snyder to birmingham where my mom's family is from.
third most is I 65 north from the kennedy bridge (yes I know the kennedy is a southern aligned bridge now. it wasn't always) to exit 7. lot's of trips to salem, in on top of a few trips to indy and chicago plus the start of a few west coast road trips that used 65 north.
Probably I-10 between LA 1 and I-110 in Baton Rouge if I had to guess, though I haven't been on it in 9 years. As a driver, probably I-69 on the northeast side of Houston.
I-190 in NY, because it just so happens that I use that Interstate most of the time I go to the US via NY.
The I-5 Santa Ana Freeway in southern California. Lived in OC and worked in downtown LA for a few years. Awful commute.

Hard to say given how many times I've moved. At various times my commutes have included:

Within the last 8+ years, it would likely be a tossup between I-280 between downtown San Jose and CA 85 in Cupertino and I-880 from its southern terminus north to Fremont Blvd. in south Fremont. For three years prior to that, it was definitely I-15 from Ontario to Main St. in Hesperia; and before that I-10 from Ontario to Redlands.
For me it's almost certainly the bit of I-290 IL in red, followed closely by the bit in blue. Past that, I've been on the remaining bits of I-290, I-294 and I-88 pretty much equally, except for that bit of I-294 parallel to I-290, which I can only recall traveling on three times.

Well, seeing as the second nearest I-route is about a half hour drive time away and in a direction that I don't often have a need to go....
(Before I-41 was signed, I would say that it was I-94 between the Zoo Interchange and downtown Milwaukee.)

As a kid likely interstate 55 between interstate 12 in Hammond and interstate 20 in Jackson.
As an adult: Interstate 10. Ive lived in Houston, Baton Rouge and New Orleans and been to San Antonio often so I've been on interstate 10 more than any other; though I would guess interstate 45 is a close second–actually interstate 69 between downtown and sugar land may be tied with 45.

I'm sure it's the section of I-135 between Kellogg and 53rd Street, here in Wichita. I drive it twice a day as part of my daily commute.
