For me, US-12 between Fort Atkinson, WI and Whitewater. I have driven that stretch literally thousands of times. (Which would make WI-89 the state highway I have driven the most since they duplex.)
Eesh. I commuted on US 53 from Superior to Duluth for over a year.
But, I also ended up on US 5 in western MA a whole lot living there for over a decade and then US 9 and US 9W in the Capital District.
Probably US 9W.
The portion of US 1 that's concurrent with I-95 between Dedham and Canton.
Probably the section of US 10 in Elk River, MN.
A section of US 22 in the Montgomery area (NE of Cincinnati).
I-95/US 3 east of the Burlington Mall exit.
US-50 Sacramento because I used to work and go to school in Sacramento for 4 years.
There are legitimately no US routes in my area. Probably the following sections of US 20, in roughly this order:
(1) NY 444 to US 20A (eastern terminus)
(2) NY 332 to NY 364
(3) Lake St (Geneva) to NY 96A
Either US 27 in Lansing, MI or US 1 from Cudjoe Key to Key West.
Probably US 12 in Sturgis, when visiting my wife's family before her folks passed away. Although US 23 would be a close contender, probably the winner in recent times.
US 6 in Marshall County, IN, between IN 106 and Plymouth-Goshen Trail.
US-30 between Fort Wayne and Columbia City, IN
US 41 (prior to I-41 designation) Green Bay to Menasha.
US-23 between Saginaw and Flint. Which happens to be the same answer as my interstate highway because US-23 runs concurrent with I-75 in that stretch.
For a US highway that isn't concurrent with any other highway it'd probably be US-10 between Midland and Bay City.
close call between US 74 (grew up in Charlotte) and US 70 (currently work and live along it).
It's a toss-up. I grew up in NC in Wayne County, so it's a tie between US-117 and US-70 in the whole county, as well as the part of US-117 in Wilson County.
US 101 on the San Francisco peninsula.
I use a stretch of US 441 in Gainesville on my daily commute, but it's going to take years for that to beat out any of the US Routes in my hometown.

It has to be US 20 in the Wakeman, Ohio area-specifically from Green Road to Wakeman proper and on to Western Reserve High School near Collins. Besides trips with my parents, I took that route every day on the school bus (or later driving myself) from Kindergarten to High School graduation. And I took that route often when working after college graduation until I moved to Colorado at age 28.

US-29 in Gainesville, VA. Literally can't leave my neighborhood without going on it.
2012 to date: US 101, generally between Gilroy and San Francisco.
2009-2012: US 395, southern terminus to CA 58.
1997-2009: US 101, southern terminus to as far north as I needed to go (more often than not friends in Goleta).
1992-1997: US 26 from downtown Portland out to Foster Road (then-GF living outside of Boring), or:
then-signed Business US 30/Sandy Blvd. from downtown out to about 82nd/OR 213.
Prior to that, it was all CA, and there's not been a lot from which to choose for 57 years! Guesstimate: more US 101 than anything else.
Overall for me is almost certainly US 89 near Salt Lake City, but more recently US 29/78/278 in Atlanta.

In recent history, a toss up between US-20 and US-97. The only two highways running through Bend, and I'm on them a lot.
When I lived in California, it probably was the 101.
When I lived in Wisconsin, I was on 151 through Madison a lot when I was going to school there.
I would say that it's a close race among 4 candidates:
Two sections of US 5: One between the center of Wallingford and the WC Parkway, the other as part of the Berlin Turnpike
The portion of US 6 concurrent with I-84 between Farmington and downtown Hartford
US 44 in Avon/West Hartford (went to school off of there for 11 years)
Well considering I live near US 92 and go to work using the latter, I would say 98 the most and 92 second.
US-31 in Manistee
I lived most of my years in Minnesota so highway 10s concurrency with 694 and 35E
US 101 between Post Street in San Francisco and Broadway in Burlingame. At one point I used to frequent US 50 between West Sacramento and Howe Avenue, back when I lived in Sacramento, but haven't been on there nearly as much since returning to the Bay Area.
US 2 on the Hewitt Avenue Trestle from Everett to Cavalero (where it splits with SR 204 and 20th Street). The westernmost section of the western section, a dangerous and congested expressway, and also home to some signage errors:

US 14 in the northwest suburbs of Chicago, notably between Palatine and Crystal Lake.
Pretty sure it's US 1 for me. Even though I currently live closer to both US 60 and US 360 and actually lived on US 60 before, I've driven US 1 in several states many times over.
