If you haven't clinched every interstate in your state (like me), that would be your answer. But if you've clinched them all, which part have you been on the least?
For some reason I don't hit I-710, I-605 and I-105 very often.
Haven't been on I-77 between I-81 and the West Virginia state line, I-66 between US-29 and US-17, nor I-381.
I've hit all the other interstate highways in their entirety in the state, including all of I-64, I-81, I-85, and I-95, plus the other segments of I-66 and I-77. Additionally, all of the 3di's except I-381 - I simply have no reason to drive on it.
There are a couple segments of I-469 that I've only been on once.
I-64 west of IN 69 only twice.

I'm surprised that you've made it onto I-90, I-80, and I-76 but never the Ohio Turnpike.
For me it's I-15 between Idaho Falls and Dubois, never been on that at all. In that general region I'm going to either be on US-20 or ID-22 on any road trip from Boise, unless I need to stop somewhere else along the way, which is not very likely (eg. stopping in Idaho Falls on the way to Butte for whatever reason).
Honorable mentions:
- I-15 between Pocatello and Tremonton I've only been on once, very unlikely to change (since going to Utah I'm almost always going to take I-84).
- Most of I-90 I haven't driven myself (I've been a passenger on it twice), and living in Boise that's not likely to change soon. Eg. if I am driving to Missoula I'm going to take US-12 or US-93, not I-90.
- I personally avoid I-86 when I can in favor of US-20 or I-84 (unless my destination is Pocatello or continuing on US-30), but I still end up on it on occasion (eg. avoiding winter weather on US-20 driving to Idaho Falls), I think I've been on it 3 times now (only once as the driver).

Yea only been on the free sections of those 3. The Ohio Turnpike runs in a somewhat NW-SE alignment, which isn't really helpful for someone like me living in the southwestern portion of the state. I got I-76 and I-80 by convincing my dad to use the I-71/I-76/I-80 routing instead of the standard I-70/I-78 on a trip to NYC, as I was bored of the PA turnpike at that point. For I-90, it goes in a SW-NE direction east of Cleveland, sort of like a continuation of I-71's alignment, so it helped us on a trip to Niagara Falls, and maybe Boston if we drive that far in the future.
I've been struggling to come up with a concrete answer that I'm happy with, but in general it's I-90 between Austin and the US 14/61 split near La Crescent, and I keep zeroing in specifically on the section between MN 16 and US 52. Due to better options from the north to get to destinations along/in the orbit of I-90 (US 52 to Rochester, US 61 to Winona, US 63 to Stewartville, US 218 to Austin), there's little reason for me to get on 90 unless I'm up to something specific; I've also now clinched everything in southeastern Minnesota, so there's no further reason for me to use 90 either.
I've clinched the Interstates in VT (not super hard to do), but there are three stretches I've done only once:
I-89- Exit 21 (VT 78) to Canadian border
I-91- Exit 11 (US 5/WRJ) to Exit 26 (VT 58); Exit 28 (VT 105) to Canadian border

I believe that I have driven every
section of Interstate in California that can be driven (not necessarily the last couple meters on I-5 before the Mexican frontier). I am not sure that I have clinched I-215 and I-8 while they were called by those names. I have driven the fewest number of times on the eastern sections of I-8.
I-69 between Flint and Port Huron. Close second would be I-69 between Lansing and Flint.
In Virginia, I haven't been on I-81 south of I-581 and north of the southern end of VA 37, I-64 west of US 11, or I-77.
I think the only section in MD I haven't driven myself (though I have been a passenger) is the brief <1 mile piece of I-295 heading north into DC from I-95/495 by National Harbor.
Much of I-69 from Marshall to Port Huron.
Honorable mentions (I have been there but not lately and not often):

I have driven every mile of interstate in Utah. The only segment I haven't been on more than once is the bit of I-70 over the San Rafael Swell, between US 89 in Salina and US 6 west of Green River.

If I'm answering the question based on where I have driven... only been on I-85 and I-95 between Petersburg and US-58 once, and it was specifically to clinch. I have no reason to logically drive those roads. Also I-66 between US-17 and I-81.
I-94 between Eau Claire and Elk Mound.
Never been on 76, 490, 670, 675, 680 in Ohio. 75 and 475 the most. 70, 71 and 270 several times around the east side of Columbus. Only on the others once or twice.
Been on all freeways in Michigan and Colorado
Definitely I-70 west of I-135/US 81.
granted, I may have been on this part of highway when I was younger and just don't remember, but I 24 between the western kentucky parkway and I 65 I don't recall being on. the only sections I have only been on once are I 75 between I 71 and 64, I 64 east of 75, and I 24 west of the purchase parkway. note: I am not counting I 69 as it is not connected to any other part of constructed 69 or the parkways redesignated as 3di interstates.

Never been on I-41/94 south of the Milwaukee airport. Also never been on I-43 between Beloit and I-894. And I would be hard pressed to find a real reason to drive either of those.
I-90, I-579 and I-279 in Pennsylvania -- opposite side of the state from me. I have been on the others, but just not clinched them all.
I-95 in Florida from West Palm Beach to I-195 in Miami.
Never had call to drive any of I-30 from Texarkana, AR to Benton, AR, nor I-55 north of Turrell, AR.
I-395, except for the portion concurrent with CT 2A (to go to Mohegan or Foxwoods). I think I've driven the portion north of SR 695 exactly once.
I have clinched the entire Interstate system in Maryland.
That said, the segments I have been on the least:
I-70 between I-695 and the Park-n-Ride (or has that been decommisioned?) Once.
I-270 Spur between I-270 and I-495. Once or twice.
I-395 into Baltimore (both forks). Probably five or less.
Everything else, more often than I can count.
I-24 between the Tennessee state line and the WK Parkway (I-69), and also from US 60 to the Ohio River bridge. Also, I-71 between KY 227 and US 127, and I-264 between US 31W/60 and I-64 near the Sherman Minton Bridge.
I-40 in its entirety between Barstow and Needles. That followed by I-8 east of El Centro and I-10 east of Route 86. Interestingly, another one I also can't say I've been on is I-605 between I-10 and I-210.
I've driven every single mile of Bay Area interstate and all of I-80 in California, and I've been down (as a passenger mostly) all of I-15/Route 15 and I-5. For the LA auxiliary interstates, I've been down all of 210 except for the section that is now 57, all of 405, all of 110, all of 710, and all of 105.
Up north, I've clinched most of 505 but my family and I may have gone up the north portion when I was 1 year old (on a trip to Portland and Vancouver).
I've been on I-705 in Tacoma twice in my life.
I've been on I-5 North of Bellis Fair Mall about 5 (usually take one of the State Routes to Langley/Surrey or Abbotsford, rather than Vancouver Proper.
Aside from I-5/405 around Seattle, everything else in the state is about equally traveled.
I only recently moved to SoCal from the Midwest and haven't been in No Cal since 1982. I'm pretty sure I've never been on I-505 or I-780 and I don't remember being I-980 either. (I may have to drive it soon if some OCD urbanists have their way.) I'm fairly sure I have yet to be on I-805 in San Diego. I'd have included 905, but it's a state highway. California is a big state, so I'm sure there are many places I'll never see.
Being in Northern Nevada I rarely have much reason to be on I-15 outside of the immediate Las Vegas area. My least traveled part of I-15 would be from US 93 to the Arizona border.
Of the three states I've lived in:
Louisiana: I have driven every mile of interstate in Louisiana including future interstate 49 between Lafayette and New Orleans
Mississippi: I have never driven on the new Interstate 69, interstate 269, or interstate 22
Texas:

No, I'm from Chicago, but I live in Tennessee, at least most of the time. Others have expressed confusion before as well, as seen below, but I'm still reluctant to change my location.

In Michigan, I've clinched just about every mile of interstate, save for I-94 east (north) of Mt. Clemens, and almost all of I-69.
I've done all of the I-routes here in Wisconsin, but the one that I have done the least is I-535. Second least is likely I-94 between WI 21 and US 10 and/or I-90/94 between the Tomah interchange and the I-39 Cascade interchange.

90/94 between Mauston and Tomah after never driving 535 would be next on the list. Followed by I-43 when it leaves Milwaukee by itself to head towards Beloit. Living by Adams at that time quicker to take 21 back to 13.....and regardless of heading towards Lacrosse or Eau Claire that stretch of 90/94 from Tomah towards Mauston only took if wanting to drive a different path. Have made drives to Janesville, Beloit and points on south making I-43(always WI-15 to me) drives more required.
Probably the section of I-244 on the north and west sides of downtown Tulsa. I don't go to Tulsa very often, and when I do, I don't go downtown, and when I do, I usually opt for I-444 because it's a secret interstate, which is cool.
In Wisconsin, I have only been on I-535 a handful of times.
For 2dis, it would clearly be I-94 between Eau Claire and the MN border.
In West Virginia, I-77 between Charleston and Ripley is the lowest for me followed by I-64 between Sandstone and Sam Black Church and the West Virginia Turnpike between Chelyan and North Beckley. I've driven all of these segments more than once, but living in Morgantown, there are shorter paths to get where I want to go than via these segments.
I think I've only been on I-70 between Grand Junction and the Utah line 4 times, which would be by far my least traveled stretch.

I haven't been on I-59 in GA, so that's the answer.
Tie, I-74 west of I-275 & I-480 between OH 91 & Oh Tpk. Both I have yet drive on in 46+ years.
That would have to be I-805 from CA 15 south to its terminus just north of US Customs along I-5. Most of my time in SD has been in the northern part of town; rarely got down near the border.
Definitely I-95 between I-26 and I-20. This is also undoubtedly the least crowded stretch of 95 between the Northeast and Florida. All I really miss seeing along this stretch is the bridge over Lake Marion and a bunch of trees.
Until recently, I lived in NE IL and my answers would be I-64 once it gets beyond Metro East (East SL area), and I-57 south of Dillingham. Haven't driven either one. But, now I live in WI but I've already clinched the entire interstate system here (even 535), But I'd have to say that the small segment of I-90 between Beloit and Janesville is one I've driven the least, and I haven't driven it since 2009.
And that's ironic, yet pretty typical. If you look at a map centered on your home and imagine it's a hub with roads branching out in each direction, the hardest ones for you to clinch would normally be an interstate that's just beyond your area and which runs perpendicular to that grid that I described earlier. In my case, the stretch I've least used is only 65 or 70 miles from me (compared with I-535 which is over 400) and is perpendicular to that imaginary grid. From my current location (or even my prior one in IL), to get to Madison would either be via US-12 to I-39/90, take an alternate version of this route which includes a piece of I-43 to WIS 11 to I-90 in Janesville or I-94 to I-894 to I-94 again. Never a reason to take I-90 in that stretch.
I'll bet a lot of you have a similar story in your own states.
Over the course of my life... I'm not sure I can narrow down the #1 spot.
However, somewhere right at the top of the list is probably the section of Interstate that's closest to home, less than 1½ straight-line miles from my house: I-35 between exits #45 and #50. I can only recall having been on that stretch of the Turnpike one single time. I never have any need to be on it. The only time I remember driving it was when I was out of town with friends, and we were following them somewhere else before heading home.

What's funny is in regards to my post on Post number 58 in this thread I was on I-75 between St Ignace and Sault Ste Marie yesterday. And rode over the same stretch of I-75 three times as I circled St Ignace.
For that stretch of I-75 being one of the lowest traffic counts on the interstate highway system it seemed pretty busy.

Same for me and the section of the Thruway closest to my house, as I've mentioned several times. There are 3di's that are closer, but that's the closest 2di.

Currently near Alexandria. I grew up in Shreveport so New Orleans was the far corner of LA away from me. I went to college in Monroe. I've been on I-510 but only to get to Michoud Blvd to look at a friend's house months after Hurricane Katrina. I go to Gulf Shores and the Florida panhandle almost once a year so I'm always right there at the 10/12/59 intersection but never a reason to turn north, so I'm missing I-59.

Counting California as a "your state" since I grew up there, I've been on every Interstate segment, but the part of I-5 between the border and exit 1 I've been on only a round-trip, on a high-school Spanish class field trip to a Tijuana high-school English class, would be the one I've been on least. This was in the early 1970s, when border crossings were much less painful than they are now.
As for Virginia, I-77 north of I-81 (for 2dis) and I-564 in Norfolk (for 3dis) would be my winners.
I-8 and I-40 are the freeways I never been on in my home state of California.
I have driven every mile of interstate in Nevada in my adult lifetime. But I have only done one round trip on I-80 between Elko and the Utah state line, and only two trips on I-15 between US 93 at Apex and the Arizona state line.
I-10 west of Vicksburg. Most of I-40 west of Flagstaff (excluding the concurrency with US 93, and west of there to where AZ 95 heads south to Lake Havasu City).
I have never been on I-37. I have never been to San Antonio!
I haven't been on I-14 since it was signed,
I have not been to the valley since I-69's letter parts were built / signed. So I haven't been on I-2 either.
