(In addition - because most of these will involve California or Texas, for obvious reasons - I also pose this bonus question: what are some of the longest drives, in terms of distance, between cities or towns in neighboring states east of the Mississippi River - the side of the country where states tend to be smaller? One of the longest I can think of there is from Memphis, TN to Elizabeth City, NC, which covers 915 miles. Also, Dalton, GA to Key West, FL is about the same, at 909 miles.)
You can add to the GA-FL example by using a place along I-59. For example, Key West FL to Trenton GA is 956 miles.
I imagine driving from Gravette, AR to Johnson City, TN is no mere day trip.

If we expand this to Canada, I have to think a drive from St. John's, NL, to Caniapiscau, QC, would be up there in terms of distance. I plotted a route via Labrador City in order to keep the entire trip within the two adjacent provinces. Google Maps quotes 2,096 km from St. John's to Labrador City via the Trans-Labrador Highway and 2,773 km from Labrador City to Caniapiscau, for a total of 4,869 km (equivalent to 3,025 miles).
If instead you took the route through Nova Scotia and New Brunswick, the distance would be 3,783 km via the Argentia ferry or 4,170 km via the Port-aux-Basques ferry.
I don't see a way to select shortest route on Google maps, so these are the fastest way (and I also picked "cities" that showed up, which may or may not be CDPs):

Gonna put down two states that are the big nothing.
Yeah, it clips Wyoming, but Google Maps says the drive between the gated neighborhood of Dakota Dunes, South Dakota, and a tiny town called Yaak in Montana is 1,300 miles long. You need to add 57 miles to avoid Wyoming.

If you count these as "cities", it's pretty long, but Yaak is an unincorporated community of 248 and Dakota Dunes is a gated neighborhood by North Sioux City.
For the "East of the Mississippi" version, I was able to top the 1000 mile threshold:
Key West, FL to Florence, AL is 1051 miles (per Google Maps), with a waypoint where I-10 crosses the Apalachicola River.
Without that waypoint, the suggested route goes through Georgia (does that violate the OP's rules?), and is longer to boot. I made a SWAG* that such a waypoint would be on the recommended route between those two places while constraining to the two states in question.

Red Lake, ON to Old Fort Quebec, QC: 4355 km. (It does cross north into Labrador). If you end at Kegashka, it stays in the 2 provinces for 3234 km.
Dinosaur CO to Idabel OK: 1188 miles via the fastest route through Kansas, or 1162 if you use the most direct route. There might be a longer one, but it looks like far NW Colorado is pretty devoid of actual towns.
Rodeo NM to Miami OK: 1035 miles via Texas, or 1130 miles via the Oklahoma Panhandle.
If it wasn't for Cape Cod and the far northeastern corner of Vermont, Buffalo to Montauk would be further than Buffalo to anything in any neighboring state.
I would say Scranton to Binghamton is the longest in my journey. 61 miles
Also Fredericksburg to Winchester (which is bordered by WV) -> 81 miles

Virginia borders Tennessee. Using my iPad, I'm not sure of the total distance because the Google Maps app is crap for multiple stops (won't give total distance), but Memphis to Chincoteague is over 1,000 miles sticking just to the two states.

Not quite: Memphis TN to Hatteras NC is only 998 miles. It's only 977 going up the coast to Corolla, NC. I wondered about the ferry ride to Okracoke, but the trip distance is significantly reduced using the Swan Quarter ferry instead of the Hatteras ferry.
Harpers Ferry WV to Hickman KY is only 814 miles. But most of that trip is through Virginia and Tennessee.

I didn't try to force Google Maps to cross directly from Kentucky to Virginia via one of the five logical crossings (US 421, KY/VA 160, US 23, KY/VA 80, or US 460) to see what the distance would be staying in those two states and not going into other states.
