SR 26 initially travels north from MD 353 along Millsboro Highway to Gumboro through an agricultural area west of Cypress Swamp. The state route overlaps with SR 54 from the state line to Cypress Road, and prior to Summer 2022 with SR 30 between Whitesville Road and Shaft Ox Corner. The remainder of SR 26 travels from west to east.

Following Nine Foot Road to U.S. 113 and Dagsboro, SR 26 dog legs town along a combination of Clayton and Main Streets to Vines Creek Road. SR 20 briefly combines with SR 26 on this stretch. Vines Creek Road crosses Hamplin Neck to Clarksville and a turn onto Atlantic Avenue. Atlantic Avenue continues the route east across White Neck to the towns of Millville, Ocean View and Bethany Beach.

SR 26 appears on the 1936-37 Delaware Official Highway Map along its current alignment. Growing beach traffic along the route between Millsboro and Bethany Beach led to two-year, $25 million improvement project along a 4.14 mile section of Atlantic Avenue, from Powell Farm Road east to the bridge across Assawoman Canal. A continuous center turn lane and bicycle lanes were added, new traffic lights were installed at Powell Farm Road, SR 17, Old Mill Road, Central Avenue and West Avenues, and a sidewalk was constructed from Windmill Drive to Assawoman Canal. Additionally a closed drainage system addressed a prolonged problem with street flooding along the corridor. A ribbon cutting ceremony held at the Millville Volunteer Fire Company on September 19, 2016 celebrated completion of the work.1
