Alabama 21 begins at the state line, continuing Florida 97 northward to Atmore in Escambia County. Alabama 21 and Florida 97 provide an alternate route to Pensacola, Florida in lieu of U.S. 29 and Alabama 113 to the east.
Alabama 21 provides a four-lane route southward from Interstate 65 into the city of Atmore and a two-lane rural roadway northward to Monroeville. Seemingly lost-in-time towns dot the landscape between Interstate 65 and the Monroe County seat including Uriah and Frisco City. At Frisco City, a short Truck Alabama 21 exists, bypassing the main portion of town to the east. Within the Monroeville vicinity, Alabama 21 crosses U.S. 84 and overlaps with Alabama 41 northward on a four lane alignment. Upon reaching the core of Monroeville, Alabama 21 splits between a western Business alignment and an eastern four-lane Bypass alignment. The split lasts two miles in each direction, with Alabama 47 tying into Business Alabama 21 at Downtown Monroeville.
Further north, Alabama 21 sees overlaps with U.S. 80 leading into Montgomery and with U.S. 80, 82 and 231 around Montgomery on the South and East Boulevard system. U.S. 231 and Alabama 21 combine through Wetumpka and Sylacauga, where the state route shifts to its own alignment northeast to Talladega and Anniston.
