A rural route for the most part, SR 29 travels 75.82 miles from U.S. 27 near Palmdale in Glades County to U.S. 41 at Carnestown in southern Collier County. The state road joins the city of Immokalee with I-75 (Alligator Alley) to the south and the Hendry County seat of La Belle to the north. Collier County Road 29 extends south from SR 29 and U.S. 41 another 7.87 miles to Everglades City and Chokoloskee.

What was the first northbound reassurance marker for SR 29 was cosigned as I-75 erroneously. Since corrected, SR 29 leads north 17 miles to the western of two interchanges along Alligator Alley. Immokalee is another 21 miles from there. Photo taken 12/31/03.
SR 29 south
Leaving the one-block concurrency with SR 80 west, SR 29 follows Main Street south from the La Belle city center. Felda lies 15 miles ahead, followed by Immokalee in 24 miles. Photo taken 03/03/03.
In northern Collier County, about six miles north of Immokalee, southbound SR 29 intersects the east end of SR 82. SR 82 travels west to Fort Myers in Lee County, while SR 29 continues south toward Immokalee and Everglades City. Photo taken 03/03/03.
Immokalee is located at the crossroads of SR 29 and Collier County Road 846. Meeting SR 29 (15th Street) on the north side of town is the east end of County Road 890 (Lake Trafford Road), a 2.87-mile spur to Lake Trafford. Photo taken 03/03/03.
Lake Trafford Road is shown on older maps as Collier County Road 850, perhaps with the intention that it would someday connect to CR 850 in Lee County. However, it appears that connection will not be built. As it runs currently, CR 850 ties into SR 82 to the northwest. Photo taken 03/03/03.
SR 29 leads south from Immokalee into the Big Cypress National Preserve and to a diamond interchange with Interstate 75 (Alligator Alley). Signage posted here originally referenced I-75 with east-west cardinal directions. These were later changed to the conventional north-south banners for the Interstate. Photo taken 12/28/03.
Another 17 miles to the south is the state road conclusion at U.S. 41 (Tamiami Trail). South of U.S. 41, SR 29 becomes Collier County Road 29 en route to Everglades City and Chokoloskee. Photo taken 12/28/03.
