State Road 46 travels east from U.S. 441 at Mt. Dora to U.S. 1 at Mims. A small section of Lake County 46 spurs west from U.S. 441 (Orange Blossom Trail) to Old County 441 east of Downtown Mt. Dora. The shared alignment with U.S. 17 & 92 (French Avenue) through Sanford is unsigned; instead trailblazers are posted at each end for the continuation of Florida 46.

Construction of Wekiva Parkway east from near Mt. Plymouth to Sorrento directly overlays a sections of SR 46. A portion of the route will be repurposed as a frontage road. Additionally the March 31, 2018 opening of the Mt. Dora Connector (SR 453) northwest from Orange County to SR 46 west of Sorrento, coincides with the decommissioning of SR 46 through Sorrento and Mt. Plymouth.
