State Road 616 provides access to Tampa International Airport (TPA) from Dale Mabry Highway (U.S. 92) to the east within the city of Tampa. The 1.79-mile route lines Columbus Drive and Boy Scout Boulevard west to International Plaza Mall and Spruce Street west from there to a stack interchange with both SR 60 (Memorial Highway) and George J. Bean Parkway north into the airport.

SR 616 replaced the east-west leg of SR 589, when the Veterans Expressway was completed northward in 1994. SR 589 originally followed an L-shaped route west from U.S. 92 to SR 580 (Hillsborough Avenue) on the west side of TPA airport.
