The highway overlays Historic U.S. 66 along an expressway leading into the city east from Target Creek. Following Sunset Drive, the business route navigates through the El Reno residential street grid east to Adams Park and U.S. 81 in Downtown.

Turning southward, Business Loop I-40/Historic U.S. 66 combine with U.S. 81 along Choctaw Avenue south, Wade Street east and Rock Island Avenue south. The two routes curve east through an array of industrial businesses onto SE 27th Street at Canadian Square shopping center. Historic U.S. 66 continues east from SE 27th Avenue as SH 66 while Business Loop I-40 returns to I-40/U.S. 270 via U.S. 81 south.
