New York State Route 263 travels 10.48 miles northeast from U.S. 62 (Niagara Falls Boulevard) near Eggertsville in the town of Amherst northeast to NY 78 (Transit Road) at Millersport in the town of Clarence. NY 263 follows Grover Cleveland Boulevard, a four lane arterial, northeast 0.72 miles from Niagara Falls Boulevard to a six point intersection with Longmeadow Road and Eggert Road. Continuing northeast along Millersport Highway, NY 263 expands to a five lane boulevard to the cloverleaf interchange with Interstate 290 (Youngmann Memorial Highway).
Interchanges link NY 263 (Millersport Highway) with Maple Road and John James Audubon Parkway at the campus of the University of Buffalo (UB). The state route encircles UB east to Ellicott Creek, where it narrows back into a five lane boulevard NY 263 reduces to a three lane boulevard ahead of Stahl Road and the south end of NY 270 (Campbell Boulevard) and to a two lane highway beyond Getzville to Interstate 990 and NY 78 (Transit Road).
