Spanning the width of the state, U.S. 50 enters Colorado with Interstate 70 and U.S. 6 from Green River, Utah. Overlaying the freeway east to Fruita, U.S. 50 branches south through Grand Junction, Delta and Montrose. U.S. 550 stems south from there to New Mexico while U.S. 50 turns east across the Rockies through Gunnison, Salida and Cañon City.

U.S. 50 expands into a four lane, divided highway east from Cañon City to Pueblo, where it combines with SH 96 for 14 miles before shifting south across the Arkansas River. U.S. 50 stays within the Arkansas River valley throughout southeastern Colorado, connecting La Junta, Las Animas and Lamar with Garden City, Kansas.
