U.S. 54 travels across most of the Sunflower State along a 353.07 mile long course from Liberal east to Fort Scott. The bulk of U.S. 54 in Kansas runs concurrent with U.S. 400. The two combine in Kiowa County west of Greensburg and partition east of Augusta at Pickrell Corner. The U.S. 54/400 overlap joins Pratt and Kingman with the city of Wichita.

Through Wichita, U.S. 54/400 follow Kellogg Avenue, a freeway crossing the city east from 119th Street W to K-96 and 143rd Street E. Completed east to the Kansas Turnpike in 2020, the limited access highway replaced Kellogg Avenue.

Beyond Wichita, U.S. 54 shifts northward alongside U.S. 77 to the Butler County seat of El Dorado. The two part ways with U.S. 54 resuming eastward to Eureka, Iola and Fort Scott. U.S. 54 leaves Kansas following a 0.44 mile long overlap with U.S. 69 for Nevada, Missouri.
