U.S. 63 travels across eastern Iowa north from the Missouri state line in Davis County to Minnesota at Chester. U.S. 63 joins Bloomfield with Ottumwa, where it separates with a business loop while bypassing the city along overlaps with U.S. 34 and Iowa 163. U.S. 63 and Iowa 163 run concurrently along an expressway corridor 26.6 miles northwest to Oskaloosa. Iowa 163 continues northwest to Pella and Des Moines while U.S. 63 continues to New Sharon, Montezuma and I-80 at Brooklyn.

U.S. 63 overlaps with U.S. 6 outside Brooklyn and crosses U.S. 30, 15 miles north from there at Tama. U.S. 63 shifts eastward again on the 29 mile drive into Waterloo, where it ties into the U.S. 218 freeway outside Downtown.

Northward from Waterloo, U.S. 63 upgrades to an expressway to Denver, Frederika and New Hampton. Following an eight mile overlap with U.S. 18, U.S. 63 extends 34 miles to Chester and the state line.

Through the city of Ottumwa, U.S. 63 Business branches northwest from U.S. 63 along U.S. 34 to Iowa 149 (Wapello Street) at Greater Ottumwa Park. U.S. 63 joins Iowa 149 north across the Des Moines River to 4th Street at Downtown. North from there, U.S. 63 Business/Iowa 149 follow a controlled access arterial northward to Ottumwa Industrial Park and U.S. 63/Iowa 163.

The 7.07 mile route of U.S. 63 Business in Ottumwa was approved by American Association of State Highway and Transportation Officials (AASHTO) on May 4, 2007.
