U.S. 275 passes through Fremont, Mills and Pottawattamie Counties in southwestern Iowa along a 59.2 mile course. Entering the Hawkeye State near Hamburg, U.S. 275 leaves parallel I-29 and proceeds north to Sidney, where it circumvents the city along a 3.71 mile long bypass. The American Association of State Highway and Transportation Officials (AASHTO) approved the relocation of U.S. 275 around Sidney on May 5, 2006.

Continuing north from Sidney, U.S. 275 reaches Tabor ahead of the Mills County line, and combines with U.S. 34 nine miles from there. U.S. 34/275 overlap along an expressway around Glenwood west to Interstate 29 at Exit 35. U.S. 275 turns north along the freeway to MSR 92 (Exit 47) at Council Bluffs. AASHTO signed off on the relocation of U.S. 275 west along U.S. 34 for 3.8 miles and north 12.7 miles on I-29 on October 7 2003. The former alignment follows County Road L35.

U.S. 275 and MSR 92 follow an arterial route across south Council Bluffs by Lake Manawa to the South Omaha Veterans Memorial Bridge spanning the Missouri River into Nebraska.
