U.S. 287 through Colorado travels across the High Plains from the panhandle of Oklahoma to Limon. U.S. 287 overlaps with U.S. 385 north to Lamar. Beyond a concurrency with U.S. 50 west along the Arkansas River, U.S. 287 resumes north from Wiley to Eads and northwest alongside U.S. 40 from Kit Carson to Hugo and U.S. 24 at Limon.

An unmarked overlap with Interstate 70 takes U.S. 287 west to the Denver metropolitan area. U.S. 287 accompanies Business Loop I-70/U.S. 40 west along Colfax Avenue across the city of Aurora to Downtown Denver. Federal Boulevard leads the highway north to SH 128 (120th Avenue) in Westminster.

U.S. 287/SH 128 overlap west into the city and county of Broomfield. U.S. 287 turns north at SH 121 (Wadsworth Parkway) to Lafayette, Longmont and Loveland in Larimer County. Continuing north into Fort Collins, U.S. 287 follows College Avenue to Colorado State University and Downtown. The northernmost extent of U.S. 287 runs along the Front Range toward Laramie, Wyoming.
