U.S. 400 generally overlaps with other U.S. Highways across Kansas. The route enters the Sunflower State alongside U.S. 50 east from Granada, Colorado ahead of Syracuse. U.S. 50/400 combine east to Lakin, Garden City and Dodge City.

Encircling Dodge City, U.S. 400 follows a bypass south from U.S. 50 to U.S. 56. U.S. 400 joins U.S. 56 east 6.97 miles to Fort Dodge Road east of the city center. U.S. 400 angles southeast from there to Ford and then east to join U.S. 54 at Mullinville.

U.S. 54/400 overlap east from ahead of Greensburg to Pratt, Kingham and Goddard in Sedgewick County. Continuing east across the city of Wichita, U.S. 54/400 follow Kellogg Avenue, an urban freeway from west of Wichita Eisenhower National Airport (ICT) to K-96 by the Kansas Turnpike (I-35). U.S. 54/400 continue to Andover, Augusta and Pickrell Corner.

U.S. 400 advances east from U.S. 54/77 to Leon, Greenwood County and Fredonia. Turning south to Montgomery County, U.S. 400 combines with U.S. 75 for 4.04 miles near Neodosha. Resuming an eastward course, U.S. 400 continues to Parsons, Cherokee and U.S. 69 south of Pittsburg. Overlaps with U.S. 69 take U.S. 400 south to Baxter Springs and with U.S. 160 east to the Missouri state line near Interstate 44.

U.S. 400 was established from U.S. 83 at Goodland, Kansas to Interstate 44 along U.S. 166 in Newton County, Missouri with approval by the American Association of State Highway and Transportation Officials (AASHTO) on November 12, 1994.

A great deal of local enthusiasm has been generated in Kansas and Western Missouri over the idea of assigning a new US route number to a highway corridor that runs the entire east-west distance of Kansas and slightly into Missouri. This concept will give distinction to the corridor, help facility much needed improvements, and provide the traveler with a good east-west route through the lower half of Kansas. By providing better access, it would promote tourism and enhance economic development.

An extension west to Granada, Colorado was approved by AASHTO on April 21, 1996.

This request is being made as an extension of the existing US 400, which currently ends at Garden City. By extending into eastern Colorado, the route will now become a distinctive corridor which will traverse the entire southern half of Kansas and part of Missouri and Colorado. This added distinction will help facilitate needed improvements, provide the traveler a good east-west route through southern Kansas, and promote tourism and economic development.
