
U.S. 425 travels 120.97 miles across eastern Louisiana, from the Natchez-Vidalia Bridge to Arkansas north of Bastrop. U.S. 425 overlaps with U.S. 84 west to Ferriday then with LA 15 north to Winnsboro and Mangham. LA 15 turns west beyond Archibald while U.S. 425 continues north to I-20 and the town of Rayville.
U.S. 425 turns west at Mer Rouge, joining U.S. 165/LA 2 for 7.48 miles to LA 139 in Bastrop. Following the couplet of Franklin and Washington Streets, U.S. 425 leaves Downtown Bastrop and heads northeast to Log Cabin and into Arkansas at Rawls.

Louisiana applied for the establishment of U.S. 425 south from the Arkansas state line 17 miles to U.S. 165 at Bastrop to the American Association of State Highway and Transportation Officials (AASHTO) on March 31, 1989. Action by the AASHTO Special Committee on U.S. Route Numbering was withheld. A letter dated June 13, 1989 to the Louisiana Department of Transportation & Development (LADOTD) indicated:
The Committee needs additional information on the horizontal and vertical curvature of the route, and requests that the application be resubmitted with this additional information.
LADOTD responded to AASHTO on July 27, 1989, indicating that there were no horizontal curves or any grades that exceeded AASHTO standards. The subsequent application was approved by AASHTO on October 7, 1989. The reason for establishing U.S. 425:
This route designation would enhance economic development along the corridor and serve the interstate travel between Louisiana and Arkansas
The U.S. Department of Transportation approved the southward extension of U.S. 425 from Bastrop in December 2005. The route overlaps U.S. 165/LA 2 east to Mer Rouge, and then replaced LA 133 south to Oak Ridge. U.S. 425 replaced LA 137 south from LA 133 outside Oak Ridge to Rayville and LA 15 near Archibald. U.S. 425 overlays LA 15 south to Clayton, where it replaced U.S. 65 to Ferriday and Natchez. The extension was sought to improve economic development and potentially increase highway funding to the area. New signs were ordered in advance and completion of sign installation was expected by March 2006.1
AASHTO previously approved the extension of U.S. 425 south from Bastrop to Natchez on February 17, 2005. The application cited that U.S. 425 will facilitate travel between the capitals of Arkansas and Louisiana. It also included eliminating U.S. 65 between Natchez, Mississippi and Clayton
