Originating in Georgetown, U.S. 701 travels 61.67 miles through the Palmetto State northward to Conway, Loris and Tabor City, North Carolina. Historically the highway continued southwest from U.S. 521 at Georgetown alongside U.S. 17 to U.S. 52 in Charleston. Deemed redundant, the overlap was dropped with approval by the American Association of State Highway and Transportation Officials (AASHTO) on June 15, 1992:
The approval of this application will enable the Department to simplify the signing along this section and thus provide clearer directions for motorists traveling between Georgetown and Charleston.

The first shield for U.S. 701 posted northbound stands beyond the wye intersection with Black River Road (Road 82) in George Town.
