Virginia Route 33 extends 72.74 miles east from U.S. 33 in the capital city of Richmond to Stingray Point along Chesapeake Bay.

Within the city of Richmond, SR 33 follows the couplet of Hancock and Harrison Streets four blocks north from U.S. 250 (Broad Street) to Leigh Street in the Carver neighborhood. SR 33 parallels U.S. 250 and I-64/95 southeast to the Martin Luther King Bridge across the Upper Shockhoe Valley neighborhood. SR 33 navigates along Mosby Street, Fairmount Avenue and Nine Mile Road through the East End neighborhood to the Henrico County line at Interstate 64.

SR 33 loops north from I-64 along Nine Mile Road through Highland Springs. The state route shifts south of the freeway near Sandston, joining U.S. 60/SR 156 at Seven Pines. U.S. 60/SR 33-156 (Williamsburg Road) overlap 3.64 miles east to Elko Road, which takes SR 156 south toward Hopewell. SR 3 remains alongside U.S. 60 east 2.76 miles to Bottoms Bridge and into western New Kent County.

Turning 0.22 miles north from U.S. 60 (Pocahontas Trail), SR 33 (New Kent Highway) shifts onto Interstate 64. SR 249 follows the pre-freeway alignment along New Kent Highway east to Quinton and New Kent while SR 33 overlaps with I-64 between Exit 205 and 220. SR 33 branches northeast along a four lane highway to West Point, SR 14 at Shacklefords and U.S. 17 at Glenns.

SR 33 combines with U.S. 17 (George Washington Memorial Highway) north to Saluuda in Middlesex County and then along U.S. 17 Business (Gloucester Road) 0.80 miles northeast to General Puller Highway. SR 33 resumes an easterly course with four lanes to Harmony Village, where the state route combines with SR 33 to Hartfield. SR 33 (General Puller Highway) spurs east from SR 3 (Twiggs Ferry Road) 8.41 miles to Deltaville and Chesapeake Boulevard at the community of Stingray Point.
