Put up 2 freeways and next person who posts picks one and then puts up 2 more and on and on it goes. Can even be the same freeway through different cities. Criteria can be access to amenities, attractions, aesthetics like skyline views, freeway design. I'll start. I-35 through Austin or I-65 through Nashville?
I say 65 in Nashville.
I-70 between Denver and Grand Junction or US-101 between Santa Barbara and L.A.? (No, I don't feel bad about taking arguably the two most iconic right off the bat.)

Good choice. I-70 through St. Louis is pretty fine, especially with the new bridge. I-70 through Kansas City is horrid, outdated design.
Okay, I'll go with the obvious on this round (and they happen to be my two favorite roads). Pennsylvania Turnpike or New Jersey Turnpike?
I prefer the Virgin River Canyon on I-15 to I-70 east of Glenwood Springs, but both are pretty spectacular. My favorite city view is the coming into St Louis on the PSB with the Arch in front of the St Louis skyline, but I'm biased since I lived there almost 30 years and the view meant I was almost home.

Wow, we are really

Briley Pky or Ellington Pky (Nashville)? (better relief route?)
TN 155/Briley Parkway for sure. I've never even used Ellington Parkway, but I'm not sure why I would, unless I was going somewhere near it in Nashville. Meanwhile, the northern half of TN 155 is a very useful bypass, especially the northeastern section, which I would absolutely use every time to get from I-24 or I-65 to I-40 and vice-versa, unless I wanted or needed to go through downtown for some reason.
Okay, here's one that comes with an honest question: I-44 through Tulsa or OK 364/Creek Turnpike around Tulsa? I-44 is much shorter and doesn't have a toll between the two ends of the Creek Turnpike, but the Creek Turnpike has markedly better roadway design quality. Both have poorly designed signs because it's Oklahoma. The Creek Turnpike's control cities indicate it as the preferred route for through traffic headed toward Joplin in the eastbound direction, and Oklahoma City in the westbound direction. But here's where the big question comes in: Has the Creek Turnpike ever saved anyone any time compared to I-44?
Choose whichever you like for whatever reason you like, but if anyone has an answer about the time difference, I'd love to know.

I use Briley Pkwy to get to BNA coming down I-24 if I fly out of BNA instead of STL. Even if I were continuing on I-24, I'd seriously consider using Briley instead of driving thru downtown. Whether I'd use I-40 back to I-24 or just take Briley all the way to I-24, which does involve a couple traffic lights, is up to current conditions

I'll admit to sidetracking from the topic, but-- Most of those signalized intersections on TN 155/Briley Parkway between I-24 and I-40 on the southeastern side, by the airport, are what I call missed opportunities for jughandles. Think how much time could be saved for Briley Parkway traffic if they just got rid of the left turns from Briley Parkway and substituted them with jughandles and turnarounds wherever there's enough space to build them (and there actually is enough empty land to build most of them, and asking people to use the SPUI for a turnaround wouldn't be unreasonable). Before someone says "people in Tennessee wouldn't understand jughandles," I'll remind everyone of this: https://goo.gl/maps/6WXBRp3S4SQa8HXp6 . Sometimes I just feel like New Jersey, Michigan, half of Pennsylvania, and St. Louis County, Missouri are about the only places that really get the concept, and everywhere else, the occasional jughandle or Michigan Left is just a one-off experiment. (Okay, I've made my point, and I'll shut up now.)

Umm. Lake Shore Dr has iconic shots of the skyline coming from either side of town, the Museum Campus and Soldier Field. What does I-376 have? This is what I call a bad take my friend.
I-376 has views of the Pittsburgh skyline. Every city with at least a quarter of a million people has some sort of skyline. So what? I-376 also has mountains. Illinois has flatness. And worst of all, does Chicago even know what it's doing with traffic engineering? Their roads are such a mess, I'd rather stick to driving the Tri-State Tollway and anything outside of that semi-circle, because once you get inside, it's traffic hell and crap engineering. PennDOT is actually pretty good with traffic engineering; their only problem is their roads are so old, many of them don't meet contemporary standards, but at least they acknowledge it and try to fix it where they can.
So, I stand by my "bad" take. And, aside from traffic engineering, I really don't have anything against Chicago. I just have a lot more love for Pennsylvania than Illinois.
Anyway, nobody answered mine:

So, I stand by my "bad" take. And, aside from traffic engineering, I really don't have anything against Chicago. I just have a lot more love for Pennsylvania than Illinois.
We can disagree on your choice, but your reasoning is awful. First, yes, every sizeable city has a skyline. But you have Wichita, then you have Pittsburgh, then you have Chicago. Chicago is a world-class skyline. Pittsburgh is not. So "every city has a skyline" is not a reason. Second, did you consider that Chicago's traffic might be worse than Pittsburgh's because it's a metropolis of 9.5 million people compared to 2.4 million? Third, "Pennsylvaina good, Illinois bad" is not a relevant reason in the slightest to a discussion strictly about comparing two highways.
Overall, it sounds like you've either
a. Never been to Chicago (which is fine, but if that's the case then don't act like you're an expert on it)
b. Really love Pennsylvania for no justifiable reason.

Why do I have to care about skylines all of a sudden?
As for Lake Shore Drive in particular: (1) There's no good reason not to use overhead signage for this series of closely-spaced diamonds: https://goo.gl/maps/xmYCSDFDJusPtgDV6 , and the NB US 41 TOTSO: https://goo.gl/maps/e5NdavupH7kk4yJK6 . (2) The speed limit is 40 MPH on a freeway with four lanes in each direction. Is that even
close to the 85th percentile speed? Meanwhile: https://goo.gl/maps/EDcXQPtETTT2idLx9
(Added this as an edit because subjective reasons for preferring one freeway over another have turned into a thesis defense all of a sudden.)

Pennsylvania has my favorite scenery out of the entire continental U.S., and PennDOT is actually pretty good with traffic engineering. Like I said, the only downside to Pennsylvania's roads is some of them are so old, they're way behind contemporary standards, and you can't just fix all of them overnight.

I-30 in Arkansas. Honestly, I-44 in Missouri is pretty high in overall quality, and it has a lot of very scenic parts, but it also sometimes feels like it drags on forever. I-30 through Arkansas seems quick and nicely wooded, but I'll admit to not having driven the part between I-40 and I-440.

Indiana Toll Road. As tempted as I'd be to pick the Garden State Parkway, it loses points for having had some at-grades on its southern end that have been converted to interchanges, and it seems to run endlessly along the coast. At least the Toll Road connects to the Chicago Skyway and links the Windy City to Toledo and Cleveland while carrying all of I-90 and most of I-80 along the way.

I don't know about time, but the turnpike used to save me pants back in the late 2000s before I-44 was upgraded through Tulsa. With the crappy 50s design on Skelly Drive, there were some scary moments. Now that 44's been expanded and upgraded through the worst parts, it's not worth it as much to bypass it.

I just thought of a tough one! I-70 from Ohio to Maryland, through Pennsylvania, including Breezewood and the sub-standard section between Washington, PA and New Stanton.
Or I-70 to I-79 to I-68 and back to I-70, which avoids the Pennsylvania Turnpike tolls and the traffic signals in Breezewood. Frankly, I don't care whether you follow I-70 through Wheeling or use I-470. But here's the big catch:
You're driving a tractor-trailer hauling a 40,000-pound load, putting your overall weight close to the max. But you're aren't carrying bulk HazMat, which means you
are allowed to use the PA Turnpike tunnels,
and you drive for a company that pays for all of your tolls.
Choose wisely! :D

I think both large-fleet trucking companies and independent owner-operators have already voted on this one, continuing to prefer the gentler grades (and therefore, lower fuel consumption) of the Turnpike over the free I-68 alternative. The New Stanton Interchange is routinely the second busiest entry and exit point for commercial trucks in the entire Pennsylvania Turnpike system–behind only Mid County.

I live pretty close to I-495 (Capital Beltway) in Maryland, but am quite familiar with I-405 (San Diego Freeway).
I would have to go with I-405, because there is more interesting things to see from the freeway. The only large development to be seen from I-495 is Tysons Corner in Fairfax County, Virginia.
There is nothing that resembles the Sepulveda Pass on I-495. Nor is there an LAX, an interchange like the junction of I-405 and I-105. The Springfield, Virginia interchange (junction I-495, I-395, I-95 and VA-644) compares well with the southern end of I-405 at Irvine, where it merges with I-5, for size and scale, though I think the flyover ramps are higher at Springfield.

Definitely the Katy. It's the widest highway in the world, I believe, plus it has nicer-looking stacks and more scenery (if you can call tall office buildings scenery) than the Eastex. The Eastex is basically just suburban sprawl until you reach Splendora, where it's not really the Eastex anymore.
If you meant the East Freeway (which I think you might have), then the Katy Freeway still beats it out, as well.

Yes, I did mean the East Freeway; should've known that the Eastex is actually I-69/US 59.

I-55 in Missouri. Missouri does a better job with it for the most part.
I-70 between the I-470/Kansas Turnpike merge east of Topeka and the Mississippi River, or, I-70 between the Mississippi River and the Indiana/Ohio state line? I gave you two segments of close to equal distance for that one. I'm already placing a mental bet on what gets chosen, but maybe someone will surprise me.
70 between the Mississippi and the IN/OH border
Two non-interstate freeway options for NC........US-421 between Wilkesboro and Colfax, or US-29/70 between Lexington and Greensboro?
US 421 for sure.
Moving on to tollways leading into Chicago from the west: Ronald Reagan or Jane Addams?
Jane Addams because it feels like a very gradual buildup to the city. You're in the cornfields, then the outer suburbs, then the office parks of Elgin and Schaumburg, then the craziness of O'Hare.

The Reagan has very similar build up, from the cornfields out near DeKalb, to the outer suburbs, Office parks starting in Naperville thru Oak Brook including the side-by-side Tollways interchange with I-355, into the craziness that is Oak Brook thru Hillside with the 88/290/294 interchange
All that said, I would still give it to the Addams. Since they did the rebuild/expansion and added the "smart road" elements, it is quite a nice ride
I have no opinion on El Paso/I-10 and Memphis/I-40
Agree on the Jane Addams. Best road I have driven... Lighted from OHare to Rockford.

How about Lake Shore Drive in Chicago (not technically a freeway I guess but close enough) vs I-376 in Pittsburgh?
I-376 in Pittsburgh. No second thoughts on that one.
Don't forget the fact that Lake Shore Dr has a little something called Lake Michigan right freaking next to it. And it goes through Grant Park. And traffic south of I-55 and north of the Oak St Curve moves very nicely. Thank you. Don't get me wrong. Pennsylvania is a beautiful state. But there are some great gems in Illinois and LSD is one of them.
Most of I-15 in Utah/Salt Lake counties is my nomination for best flowing urban freeway. Oh if only PDX had such a great setup for I-5....

Okay, I'm going back to one of the choices in the original post, but I'm using it to pose the eternal dilemma:
Suffer through traffic on I-35 through Austin, or pay the tolls for TX 45 and 130 around Austin? I'll make it harder: pretend you miss the sign that gives the travel times.

Eh, who doesn't like to go 85 mph legally? 130 it is. TBH though, 130's surroundings are fairly interesting, plus the road is actually designed for 85 mph. Fun fact: I drove part of 130 before I had my driver's license (I only had my permit).

As a non-Texan, I wouldn't want to deal with TxTag or however that toll system works. My family once drove on the OC toll roads in Socal and the website (thetollroads.com) wouldn't let us pay online at all. How does 130/45 work exactly?

When I drove 130 a couple years ago, my family didn't have a TxTag, but we did have an EZTag (basically Houston's toll tag system). The way the EZTag works is you type in your license plate number on its app and for a week you can use the electronic toll lanes around Houston without getting fined. After the one week, the plate number expires and you get the toll bill, which you pay with a credit card. Then you can type in your plate number again for the next week.
I believe the app is free, though, so I bet non-Texans can use it without issue if they make an account. 130 said it accepted TxTag only, but after we drove it, the toll charge for the TxTag appeared on the EZTag account, so I think the systems are interchangeable. Not sure about other states' tags, though.
I have a PikePass (OK), so TX-130 just charged to that like any other toll road in Oklahoma.

Strongly disagree here. 75 through Florida is surprisingly boring. Just flat, straight, and trees. You pass by a couple metros but see nothing. Alligator Alley is super boring too, 41 is way more fun in crossing the Everglades. 75 in northern GA is nice, and going through downtown ATL is one of the best urban freeways in the country (though I've always done it at night and never hit traffic).
