Well, I suppose, but Alton IL is the one everyone knows, so I'm not sure this matches the spirit of the thread.  Do any of the other Altons even top 1000 population?

A slightly better one for Illinois would be Marion (not OH) and Harrisburg (not PA).  If only the spelling were the same, the third destination would also qualify.

If only the road between Eldorado and IL-34 weren't signed for Eldorado from IL-34, there would also be Raleigh/Harrisburg.  But alas, Raleigh is only signed from the other endpoint.

Some runners-up:
Virginia (not the state), Jacksonville (not FL)
Danville (not VA), Paris (not TX)
Stockton (not CA), Freeport (not NY)

I think it's more interesting when the two faraway locations are ones that could reasonably appear on the same sign.

I'm pretty sure that can find a sign on I-80 near Salt Lake that shows Reno and Cheyenne (opposite directions), but I'm late to the party. That probably wasn't what OP had in mind. There's definitely a "Mecca" and "Baghdad" distance sign (on US-395, if memory serves) in California.

Yeah, I've been avoiding foreign location names, because the OP specifically says "in another state".  Alternate definitions aside, I don't think he had foreign states in mind.  Otherwise, of course, we'd have Ottawa and Peru.

(I once had a co-worker with a learning disability who mentioned every so often that she was born in Peru.  Because this was in the Chicago suburbs, a bunch of us at work simply assumed she was actually born in Peru IL but was confused and thought she was born in the South American country of the same name.  Years went by before we learned her parents were missionaries.)

One Mother's Day maybe 15 or 20 years ago, we took my mom to dinner at the Ashby Inn, a restaurant in a small town in Virginia. We told her that if any of her colleagues asked, she should say we took her to Paris for dinner. (The small town is Paris, Virginia.)

(Marion PA is somewhat close to Harrisburg, but I haven't been able to find a sign with both place names on it.)

This one doesn't work, but it amuses me that, from the I-135/US-54 interchange in Wichita, you can either go to Newton or take Kellogg to Goddard.

Williamstown and Cambridge are on opposite ends of MA 2, although Cambridge is overshadowed by Boston from that direction.

I don't recall seeing a population requirement earlier in this thread  :sombrero:

But per Wikipedia, Alton, Texas has 18,198.  Memphis, Texas has 2,048.

How's this for irony? These two towns are located in a state that uses a diamond state highway shield, but they're not in the one you'd expect:

Go Cubs Go! Go Cubs Go! Hey Chicago, what do you say? The Cubs are gonna win today!

And my first thought was, "Yeah, what about it?"

I guess I should have thought of that major city, spelled the same but pronounced differently.

But when I thought Lowell, the one that immediately came to mind was in MA. I didn't know there was one in NC.

Springfield and Ludlow are next to each other in Massachusetts, but this sign is about 85 miles north in Vermont.

And the reverse... here's a sign for two Vermont-named towns in Massachusetts.

Up until 1989, this sign had three Massachusetts town names on it despite being in Vermont. Now technically it's only two, since Manchester, MA officially changed its name to "Manchester-by-the-Sea." (If only Ludlow was on this one too.)

Norwalk and Ontario, but not in southern California. The third place name on there isn't the best-known of its name either, but the famous one isn't a city.

Is that because of your hockey affiliation or the California one is just inferior to the province? Either way, power to you.

It's because on Wikipedia, "Ontario, CA" redirected to the city for some reason for the longest time in a textbook example of Americentrism.
