US 80 is mine because I've lived on it all my life.  I've even been to the eastern terminus, Tybee Island, GA.

My favorite state route--LA 1. Driven about half of it so far.  Want to drive the southern half soon.
Runnin' roads and polishin' rails.

My favorites: I-39 in Wisconsin
US 41 in the Keweenaw Peninsula of Michigan
                   Wis 55 north of Shawano

My favorites:
I-10 from Baton Rouge to Mobile
I-45 from TX-288 in Houston to Galveston
US 59 from I-610 to BW-8 in Houston
TX-288 from US 59 to I-610 in Houston

I love any freeway that is flat and surrounded by a mixture of swamp, palm trees and oil refineries .

Darkchylde

Interstates: I-70 between Denver and Grand Junction. Hell to drive, scenery is second to none. Take during fall/winter for maximum epic.
I-80 in Nevada. Less challenging, but the scenery is still good. Bring a gallon of 50/50. Your car will thank you later.
I-275 (Florida). No comment needed if you've used it. If not, go there. Now.

US Highways: US 50 between Montrose, CO and Pueblo, CO. Same reasons as I-70. Mind-blowing scenery in fall or spring, and an extremely challenging drive. Make sure your brakes work, you will need them.
US 90 along the Mississippi Gulf Coast. Take it in something that can handle in order to get the best experience.
US 287 between Amarillo and Wichita Falls, TX. Wide open, just like your throttle will no doubt be. When you absolutely, positively need to get away from the stress of the world, go here.
US 93 between I-15 and I-40, NV/AZ. More amazing scenery. Yay.
US 69/96/287 south of I-10, Port Arthur/Beaumont, TX. Not so scenic, but a prime example of the awesomeness that is Texas freeway design. It's a short stretch, but your heart will still be pounding at the end.

State highways: LA 1. Being representative of a cross-section of the entire state is a good thing. The southernmost section is the best, in my eyes. Catch it before it's gone.
MS 13 between I-59 and US 98. Take it at night - pure awesome.
CO 62. Short, but the most challenging drive I've ever tried.

I-80 in PA.  A moderate number of trucks and few cars plus the highway goes through some relatively remote tree-covered areas.  Plus, the fact that the EB and WB lanes can be up to a mile apart!

PA 42 from Catawissa to Centralia, PA.  It's like riding a roller coaster in your car!

I-59 from Gadsen, AL to I-24 and I-71 from Louisville to Cincinnati.  These sections have similar characteristics to I-80 in PA.

US 431 South going uphill out of Huntsville, AL.

Leiper's Creek Road, Williamson County, TN.  Very pretty scenery, million dollar homes and little traffic.

M-37: Best way from Gd. Rapids to Traverse City.
CR 375 in Florida.

Riverside Frwy

Interstate: Interstate 210 in Southern California.
US Route: US 395 with US 101 coming in close second.
State Route Freeway:California 91(DUH :-D)
State Route Road:California 1 with California 49 in close second.
Toll Road: California 73(The alignment of this route absolute genius)

UptownRoadGeek

I-10 -> SH 6 (Texas) to I-110 (Mississippi)
U.S. 90 -> Houston to Biloxi
SH 288 (Texas) -> U.S. 59 to Pearland

Tarkus

Interstate: I-84 (the western one).  The Columbia Gorge stretch is amazing.
US Route: US-97 (my all-time favorite highway of any kind), with US-197 and US-26 close behind.
State Highways: OR-99W (learned to drive on it, so it has sentimental value), followed by WA-14.
County Roads: Evergreen Road/Parkway in Washington County, OR.  Well-built surface arterial and not too overcrowded.

Interstates: 5 (entire route), 80 from SF to Sacto, 84 (OR), 205 (OR-WA), 238 (yes, that one), 405 Portland (since it seems to be the only 405 that's not jammed throughout the entire day), 710

US Highways: 101, 99 (and all states routes associated with it), 199, 97 through Bend, 395 from the Tri-Cities to OR 140, 50 from Sacto to Lake Tahoe, 30, 26, 20

State Routes: OR 8 & 10 (having grown up near where they almost intersect), OR 66 & 140 (as part of the Oregon Outback Scenic route), OR 213, CA 1, CA 22, CA 133, CA 299 (an amazing drive to Arcata), WA 14, WA 500, WA 503

Of the Interstate's I've been on, my favorites were I-24 between Nashville and Chattenooga, and I-40 between Asheville and Tennessee (where the rockslide is).

As for non-Interstate roads, my favorite is hands-down US1 between Miami and Key West.

Hi!

Kacie Jane

Keeping this within Washington -- I may have other favorites that aren't really coming to me right now --

Favorite interstate: I-5 between about 232 and 250 through the Chuckanut Mountains... provided I'm not trying to get to Tacoma on Christmas Eve through 3 inches of snow.

Favorite US Highway: US 12 through White Pass - really the entire section from I-5 to Yakima, or 101 around the peninsula

Favorite State Highways: 11 and 542.  9 is pretty nice too.

I-295 in NJ from DE to I-95 at Trenton. Only part i really hate is the crappy junction with 76/42. Just a nice freeway to take to go south.
US-206 in NJ, Columbus NJ to Hammonton NJ, beautiful scenery when it goes through farms and the pine barrens
NJ70/72 Same thing as US-206, plus the fact that 70 has four traffic circles within miles of each other, fun to drive around.
County Route: NJ CR 537, goes from industrial to agricultural, and everything inbetween as it crosses the state.
Toll Road: Garden State Parkway from NJ72 all the way to the end in cape may county. Beautifully desegined four lane highway imho.
