Traveling east for 23.20 miles, CR 340 crosses the Suwannee River by Rock Bluff Spring into Gilchrist County and ending at U.S. 27/41 in High Springs. A separate 3.82 mile long branch along NW 156th Avenue links CR 231 with CR 225 at Monteocha north of Gainesville.
