County Road 25A follows sections of old U.S. highways between Leesburg and Lake City. Within Lake County, CR 25A comprises a 1.59 mile long loop west from U.S. 27 at Lake Harris within the city of Leesburg. The northern branch of Lake County Road 25A follows Dixie Avenue 1.95 miles to the west of U.S. 27/441 through the city of Fruitland Park.

Within Marion County, a 0.9 mile long branch of CR 25A links U.S. 27/301/441 in the city of Belleview with CR 25 to the northeast. The main leg of CR 25A in Marion County branches west from U.S. 301/441 along Old Gainesville Road in Ocala. Traveling 12.98 miles north, CR 25A connects with SR 326 near Zuber en route to Martin and CR 329 near Lowell. CR 25A returns to U.S. 441 north from the town of Reddick. This section of CR 25A was part of U.S. 441 until 1965, when the four lane highway to the east opened to traffic. The Florida State Road Department requested that old U.S. 441 be redesignated as U.S. 441 Alternate in the application sent to the American Association of State Highway Officials (AASHO) for the relocation of U.S. 441. While the realignment of U.S. 441 was approved on October 2, 1965, the request for the establishment of U.S. 441 Alternate was not. Instead correspondence from AASHTO to the State Road Department on October 25, 1965 suggested marking the former route as "Old 441".

An additional section of CR 25A loops 1.53 miles north from U.S. 441 through the community of Hague and portions of east Alachua in Alachua County. CR 25A follows NW 120th Lane to NW 59th Drive and NW 126th Avenue from there west back to U.S. 441.

The northern leg of CR 25A angles 3.11 miles northwest from U.S. 441 at Five Points in lake City to U.S. 41 near Interstate 10 at Exit 301. This branch follows old U.S. 41.
