Sumter County Road 478 forms an 11.35 mile long loop to Webster east from U.S. 301 near St. Catherine and southwest from CR 48 at the city of Center Hill. Within the city of Webster, an implied 0.35 mile long overlap with SR 471 (Market Boulevard) connects CR 478 on NW 4th Avenue with the continuation of CR 478 along SE 1st Avenue. The rural highway parallels part of an abandoned Atlantic Coast Railroad line near Matchett Lake.

County Road 478 west from SR 471 in Webster to U.S. 301 is part of the Scenic Sumter Heritage Byway.

Lake County Road 478 constitutes a 7.95 mile long route between SR 19 and U.S. 27 through portions of Groveland and Minneola. Following Cherry Lake Road, CR 478 runs through agricultural areas and by the Cherry Lake Landing subdivision northeast from SR 19 to the north side of Cherry Lake. East from Cherry Lake, CR 478 becomes more suburban with subdivisions along both sides of the route to Apshawa Road along the Minneola city line. Apshawa leads the route north and than east to U.S. 27.
