SR 35 represents the state road component of U.S. 17 north from Punta Gorda to Bartow. It overlays U.S. 98 and unsigned SR 700 northwest through Lakeland to their merge with U.S. 301 at Dade City. SR 35 accompanies U.S. 301 north from the separation with U.S. 98/SR 700 to Ridge Manor, Bushnell, Sumterville, Wildwood and Belleview. U.S. 301 combines with U.S. 27/301, and unsigned SR 25/200/500 northwest from Belleview to Ocala.

An 10.84 mile long stand alone section of SR 35 emerges just east from there at the intersection joining SR 25 (Hames Road) and CR 25 (SE 110th Street) with Baseline Road. Following SE 58th Avenue north, SR 35 (Baseline Road) bee lines to Huntington through the eastern Ocala suburbs. The state road navigates through an S-curve between Silver Springs Conservation Area and Silver Springs State Park to end at SR 40 opposite from CR 35 (NE 55th Avenue) at the community of Silver Springs.

Marion County Road 35 extends 4.95 miles north from SR 35 along NE 55th Avenue, NE 55th Avenue Road and NE 58th Avenue to Indian Lake State Forest. CR 35 concludes at a rural intersection with NE 97th Street Road west and NE 90th Street Road east.
