State Road 73 travels 49.27 miles overall from SR 71 at Chipola Park in southern Calhoun County to U.S. 231 north of Cottondale in Jackson County. The mostly rural highway is an alternate to SR 71 north to Clarksville and Marianna. Beyond an overlap with U.S. 90 west from Marianna, SR 73 provides a cutoff north to U.S. 231.

The previous shield assembly for U.S. 90 on SR 73 south. The gas station beyond the roadway split for U.S. 90 went out of business by 2013 and was subsequently demolished.
SR 73 combines with U.S. 90, 2.01 miles east to Jefferson Street (SR 166) in Downtown Marianna.

Clarksville, where SR 73 meets SR 20, is 25 miles south of Quincy. Wewahitchka lies further south along SR 71.
Unmarked here, a branch of County Road 167 follows South Street 1.75 miles west from SR 73 (Jefferson Street) to SR 276 (Penn Avenue) in Marianna.
CR 280A (Fillmore Drive) serves a handful of residential streets and agricultural areas west of the Chipola River.
Since removed, the shield for CR 280 on SR 73 south only referenced the route heading east along Magnolia Road. Thompson Road also leads CR 280 west to CR 167 (Fairview Road).
