Business Loop I-25 branches west from I-25 at Exit 298 along Main Street. The route enters the Buffalo business district and combines with U.S. 16 at Fort Street. The tandem overlap for 0.4 miles to Hart Street, which leads U.S. 16 east alongside Business Loop I-90 to I-25 at Exit 299 and I-90 at Exit 58.
Continuing north from Hart Street, Business Loop I-25 & I-90 run concurrently with U.S. 87 Business to a half diamond interchange (Exit 56A) with Interstate 90. Business Loop I-25 is acknowledged on I-90 eastbound guide signs, despite extending further north than its adjacent parent route.
