South Carolina Highway 46 runs 17.33 miles east from U.S. 321 (Deerfield Road) at Hardeeville in Jasper County to U.S. 278 (Fording Island Road) at Bluffton in Beaufort County.
S.C. 46 follows Main Street through the Hardeeville city center southeast onto Plantation Drive. Turning east along Okatie Highway, S.C. 46 overlaps with S.C. 170 for 2.37 miles to a roundabout with Mary River Road and New Riverside Road south into the New Riverside development. S.C. 170 (Okatie Highway) turns northeast while S.C. 46 lines May River Road east from Pritchardville into the town of Bluffton. The eastern leg of S.C. 46 takes Bluffton Road north from the Bluffton town center to U.S. 278 across from Waterford Drive into The Crescent golf course community.

S.C. 46 (Church Road) enters the Hardeeville city limits just ahead of Sanders Road (Road 292) and an at-grade crossing with the CSX Railroad.
