North from Interstate 35, U.S. 75 passes east of Melvern Lake en route to the Osage County seat of Lyndon. The highway bee lines north to U.S. 56 ahead of Carbondale.

Through Topeka, U.S. 75 combines with Interstate 470 west from the South / Southeast area 5.34 miles to I-70. U.S. 75 then shifts east alongside I-70-U.S. 40/K-4, 1.98 miles to the freeway spanning the Kansas River into north Topeka. Formerly designated U.S. 75 Bypass, these overlaps were approved by the American Association of State Highway and Transportation Officials (AASHTO) along with the westward realignment of U.S. 75 onto the freeway from SW Topeka Boulevard near SW 93rd Street to I-470 on April 19, 1998.

Beyond the Jackson County seat of Holton, U.S. 75 continues to Netawaka and U.S. 36 at Fairview. U.S. 75 bypasses Sabetha to the east ahead of Pony Creek Lake and the Nebraska state line.

S High Street and E 4th Avenue through the city of Caney were previously part of U.S. 75. U.S. 75 was realigned to the east due to construction of Copan Reservoir in Oklahoma. The American Association of State Highway and Transportation Officials (AASHTO), approved the relocation on June 28, 1978.

Through the city of Altoona, U.S. 75 was relocated onto a two lane controlled access bypass east of Downtown by 1979. AASHTO approved the new alignment on October 13, 1979 and a separate request by the Kansas Department of Transportation (KDOT) for the establishment of U.S. 75 Alternate along the old route on Quincy Street with the recommendation of U.S. 75 Business.

U.S. 75 originally followed Topeka Boulevard north through the capital city to Downtown and North Topeka. The route continued along Topeka Boulevard to NW 94th Street west along the Jackson County line. U.S. 75 then lined South Road north onto Eastern Avenue in the city of Hoyt. The historic route continued along 4th Street west and K-214 north to the expressway along U.S. 75. With the completion of the expressway south from Hoyt, the American Association of State Highway Officials (AASHO) approved the relocation of U.S. 75 and the establishment of U.S. 75 Bypass along I-470 and I-70 to the west on October 2, 1965.

Through southern Woodson County, U.S. 75 was realigned to improve geometry and provide a grade separation with the parallel Union Pacific Railroad line. Available for traffic on July 1, 1990, AASHTO concurred with the change on June 10, 1991. Two sections of the old route were renamed Oriole Road.

Just south of the Woodson County line in Wilson County, another realignment for U.S. 75 was made with approval by AASHTO on May 6, 2006. U.S. 75 and K-39 were relocated off the dam at Wilson County State Line. Their overlap was also shortened to 0.93 miles.

Prior to completion of the bypasses for Fairview and Sabetha in Brown County, U.S. 75 overlapped with U.S. 36 west from Bobcat Road to Fourmile Corners and turned north along the Nemaha County line to Sabetha. Completed by November 2005 and approved by AASHTO on May 6, 2006, the two lane controlled access expressway

also eliminates two dangerous 'tee' intersections and provides a more direct route through the area.
