U.S. 29 north and U.S. 80 east combine in Downtown Tuskegee at a square joining the two routes with SR 81 (Main Street) north to Notasulga.

Chappie James Drive forms part of a truck route for SR 81 north from U.S. 29/80 to Moton Field Municipal Airport (06A) and SR 81 south of SR 199.

Erroneous SR 80 shields were posted at one time for U.S. 80 at the intersection with Chappei James Drive (SR 81 Truck) north. U.S. 29/80 advance east from the city of Tuskgee city into Tuskegee National Forest.

Progressing east from Pleasant Hill across Tuskegee National Forest, U.S. 80 branches southeast from U.S. 29 at a parclo interchange with SR 186 west. The grade separation was constructed here in 1973.
Northward, U.S. 29 travels eight miles to combine with I-85 north through the city of Auburn. SR 186 runs 3.82 miles back to the west, connecting with I-85 at Exit 42.

U.S. 80 crosses paths with SR 51 at the rural settlement of Marvyn. The state route travels 15 miles southward to Hurtsboro and north 14 miles to Opelika via the community of Beauregard.

Russell County Road 33 angles northeast from Herring Crossroads and Huguley to connect with U.S. 80 opposite the north branch of CR 77 to Meadows Mill via Lee County Road 175.

SR 169 heads 1.23 miles south from the Lee County line to combine with U.S. 80 east through Crawford. Traveling 16.08 miles north to I-85, SR 169 provides an alternate route for U.S. 280/431 between Opelika and Phenix City.

SR 169 accompanies U.S. 80 east for 0.543 miles before resuming a southward heading 9.705 miles to U.S. 431 near Seale.

Crawford Road extends east from U.S. 80-280-431 as the former alignment of U.S. 80 to 13th Street and Downtown Phenix City. U.S. 280 east & 431 arc southeast 1.86 miles to their separation. U.S. 431 joins Phenix City with Seale and Dothan while U.S. 280 crosses the Chattahoochee River toward Fort Benning.

U.S. 80 west, U.S. 280 east and U.S. 431 north combine for 0.947 miles along the Phenix City bypass between Crawford Road (old U.S. 80) and J.R. Allen Parkway. The bypass was constructed by the mid 1960s between the Chattahoochee River and Opelika Road (CR 427).

Opelika Road branches northwest from Crawford Road (old U.S. 80) and intersects U.S. 80-280-431 just south of their separation at J.R. Allen Parkway. Opelika Road (CR 427) is the former alignment of U.S. 280/431 north to Smiths Station.

U.S. 80 joins J.R. Allen Parkway, a freeway completed in 1986 that bypasses Phenix City to the north to the north side of Columbus, Georgia at Interstate 185. U.S. 280/431 continue northwest along a four lane highway 22.37 miles to I-85 at Opelika.

U.S. 80 spans Railroad Street and a Norfolk Southern Railroad line just east of roadway stubs at the end of J.R. Allen Parkway.

The succeeding exit joins U.S. 80 with Summerville Road. The two interchanges along the Alabama portion of J.R. Allen Parkway are unnumbered; exits are numbered sequentially along the Georgia portion of the freeway.

Summerville Road meanders southward from the diamond interchange with U.S. 80 to become Broad Street in Downtown Phenix City. North of the nearby Lee County line, Summerville Road becomes CR 428 en route to Smiths Station.
