U.S. 29 travels northeast from Lower Alabama to the I-85 corridor at Auburn. The highway accompanies I-85 to Opelika, Lanett and Atlanta, Georgia. Providing part of a regional route to Interstate 65 from Northwest Florida in conjunction with SR 113, U.S. 29 joins Pensacola with Flomaton and Brewton in Escambia County.

U.S. 31 combines with U.S. 29 east from Flomaton to Brewton, where U.S. 31 turns north toward Evergreen. Beyond the Escambia County seat, U.S. 29 follows a mostly rural course from East Brewton across Conecuh National Forest to Andalusia. U.S. 29 overlaps with U.S. 84 at Andalusia before turning to Brantley where it combines with U.S. 331.

U.S. 29/331 run concurrent for 9.5 miles along a four lane, divided arterial or three to four lane boulevard. U.S. 331 continues north from their separation to Montgomery while U.S. 29 turns east again en route to Troy and Banks. U.S. 29 then arcs northward along to eventually reach Union Springs and U.S. 82.

Northward, U.S. 29 advances to Tuskegee, where it combines with U.S. 80 east for 7.4 miles to a parclo interchange with SR 186. U.S. 80 branches southward en route to Phenix City while U.S. 29 converges with Interstate 85 on the south side of Auburn. Through the Auburn and Opelika area, U.S. 29 previously connected the business districts of both cities. The route was relocated southward onto a 13.11 mile long overlap with I-85 between Exits 51 and 64. This change was approved by the American Association of State Highway and Transportation Officials (AASHTO) on April 8, 2000.

The last stretch of U.S. 29 in east central Alabama loops northeast from Interstate 85 to the city of Valley. The highway meets I-85 again at the final interchange in the state, which lines the south city limits of Lanett. U.S. 29 joins Downtown Lanett with West Point, Georgia as it spans the Chattahoochee River into the Peach State.

U.S. 29 overtook what was Alabama State Route 12 (SR 12 was renumbered to SR 15 by 1957) from Andalusia to Brewton, and historic U.S. 331 south from Flomaton to Pensacola by 1934.
