U.S. 250 southbound enters the state of Ohio with U.S. 40 eastbound from Bridgeport, Ohio to Interstate 70 Exit 0 in Wheeling. The US highway leaves Interstate 70 for a freeway of its own east of Downtown Wheeling at Exit 1B. WV 2 merges onto I-70 & U.S. 250 from Main Street at Exit 1A.
