State Road 37 travels 34.22 miles north from Duette in rural northeastern Manatee County to Downtown Lakeland. The bulk of the route north to Bradley Junction and the city of Mulberry serves phosphate mines in southwestern Polk County. SR 37 (Church Avenue) expands to four lanes at Dean Street in Mulberry. Advancing north between Medulla and Lakeland Highlands, the state road transitions into a five lane commercial boulevard. SR 37 enters the city of Lakeland along S Florida Avenue at Alamo Drive.

The Florida Department of Transportation (FDOT) Straight Line Diagram for SR 37 references Main Street as the north end. However signage for SR 37 is posted southbound from U.S. 98 along S Florida Avenue. S Florida Avenue continues north as SR 35 from Main Street (unsigned CR 600) to U.S. 98 (Bartow Road) at SR 548 (George Jenkins Boulevard). SR 35 is the unsigned counterpart of U.S. 98 northwest to U.S. 301 in Pasco County.

The Road Diet Test and Traffic Study along SR 37 (S Florida Avenue) in the city of Lakeland commenced on April 27, 2020. The $950,000, year long project reconfigured S Florida Avenue between Ariana Street and Lime Street from four lanes to a two lane boulevard with a continuous center turn lane.1
