Interstate 481 constitutes a 15 mile loop around the east side of Syracuse. The freeway provides a bypass for I-81 around Downtown Syracuse while connecting the suburbs of Dewitt, East Syracuse and Cicero. New York Route 481 extends the freeway another 19.5 miles northwest from I-81/481 at North Syracuse to S 1st Street in the city of Fulton. NY 481 continues another 12.3 miles north to NY 104 at Oswego.

The NYSDOT "Community Grid" proposal to demolish the Interstate 81 viaduct leading north into Downtown Syracuse includes realigning the route over I-481.

Originally, I-481 was numbered as Interstate 281 and comprised a route northward only as far as the New York Thruway (I-90) within the town of DeWitt. Interstate 481 was initially approved by the American Association of State Highway Officials (AASHO) on October 26, 1969. The belt route was completed to the southeast by 1973. Construction on the northeastern leg wrapped up in 1985.
