U.S. 29 through the Upstate of South Carolina parallels the Interstate 85 corridor from Anderson to Blacksburg. Crossing into the Palmetto State at Lake Hartwell, U.S. 29 travels 32.91 miles northeast through Anderson to merge with I-85 north near Piedmont. The two overlap northward 7.24 miles to I-185 outside Greenville. There U.S. 29 turns north into Greenville, overlaying the northern 2.41 miles of Interstate 185 to Henrydale Avenue (S-260).

Following Mills Avenue, U.S. 29 advances 0.95 miles north to S.C. 20 (Augusta Street) at Church Street. Church Street leads the highway 2.47 miles north to Downtown Greenville and Wade Hampton Boulevard. Wade Hampton Boulevard constitutes a commercial arterial leading U.S. 29 northeast to Bob Jones University and through the suburban areas of Wade Hampton and Taylors. The arterial forms a bypass north of Greer, Duncan, and Lyman, reconnecting U.S. 29 with I-85 outside the city of Wellford.

Interstate 85 stays north of Spartanburg while U.S. 29 (Warren H. Abernathy Highway) travels east directly into the city. A cloverleaf interchange joins U.S. 29 with I-26 by WestGate Mall. Through Downtown Spartanburg, U.S. 29 shifts north from Main Street along a bypass on St. John Street. East Main Street continues the highway from Spartanburg toward the town of Cowpens.

The 24.42 mile course of U.S. 29 through Cherokee County joins Cowpens with the city of Gaffney. U.S. 29 takes Granard Street to Downtown Gaffney, and Cherokee Avenue through East Gaffney. Crossing the Broad River, U.S. 29 progresses toward the town of Blacksburg with two lanes. The route converges with I-85 again just south of the North Carolina state line at the town of Grover.
