
U.S. 76 travels 296.65 miles across the width of South Carolina from the Chattooga River and Georgia state line in Oconee County to the North Carolina line at Gapway Swamp in northern Horry County. U.S. 76 traverses areas of Sumter National Forest southeast to Westminster in westernmost reaches of the state. The route overlaps with U.S. 123 east to Seneca and Clemson before turning south to Pendleton and Anderson.
U.S. 76 combines with U.S. 178 south into the Anderson County seat. The two also overlap with U.S. 29 Business/S.C. 81 for 0.73 miles along Murray Avenue through Downtown and with U.S. 29 for 0.85 miles east from Toxaway. U.S. 76/178 continue east to Belton and Honea Path before parting ways for Laurens and Greenwood respectively.
Through Laurens, U.S. 76 follows an arterial bypass to the north of Downtown while U.S. 78 Business lines Main Street to the city center along a 3.88 mile long course. U.S. 76 advances east to Clinton where it turns south along the I-26 corridor to Joanna, Newberry and Chapin.
Heading into the Columbia metropolitan area and Richland County, U.S. 76/176 overlap for 3.00 miles to Interstate 26. U.S. 76 shifts onto I-26 from Irmo south to St. Andrews, and I-126 east into the capital city. U.S. 76 runs concurrent with portions of U.S. 21/176/321 along Elmwood Aenue and U.S. 1/378 on Gervais Street leading away from Downtown.
Turning southeast from U.S. 1 at Waverly in Columbia, U.S. 76/378 remain paired to the freeway bypass to the north of Sumter. A second business route for U.S. 76 loops 5.91 miles south to Downtown Sumter, partly alongside U.S. 521. East from the city, U.S. 378 continues to Turbeville and Conway while U.S. 76 turns northeast to Timmonsville and Florence.
U.S. 76 runs across Florence, meeting U.S. 52 at Downtown and combining with U.S. 301 for 8.83 miles from S.C. 327 and the eastern bypass to western Marion County. U.S. 76 advances to Marion, Mullins and Nichols en route to Fair Bluff, North Carolina.
